长非编码RNA GHET1促进宫颈癌细胞的侵袭及转移的作用

摘要 目的 探究长链非编码RNA胃癌高表达转录本1(LncRNA GHET1)在宫颈癌中的表达及对宫颈癌细胞生物行为的影响。方法 用实时荧光定量聚合酶链反应(qRT-PCR)检测组织与细胞中GHET1的表达,分析GHET1表达高低与患者病理资料的关系。Hela细胞分为si-NC组(转染阴性对照)和si-GHET1组(转染si-GHET1)。用Transwell实验检测细胞侵袭能力,用划痕实验检测细胞转移能力,用蛋白质印迹(Western blot)法检测细胞中蛋白表达水平。结果 相比于癌旁正常组织GHET1在宫颈癌组织和细胞中升高(1.02±0.09 vs 2.96±0.26);与GHET1低表达相比,GHET1高表达与患者高国际妇产科联盟(FIGO)分级(Ⅲ+Ⅳ)、高肌层浸润深度以及淋巴结转移相关。si-NC组和si-GHET1组细胞GHET1的表达水平分别为1.00±0.13和0.46±0.10,细胞穿膜数分别为(87.53±12.53)和(32.56±8.96)个,细胞划痕距离分别为(7.15±1.11)和(3.02±0.86)μm,磷酸化哺乳动物雷帕霉素靶点(p-mTOR)相对表达水平分别为0.99±0.09和0.49±0.09,上述指标,2组比较,差异均有统计学意义(均P<0.05)。结论 GHET1在宫颈癌组织和细胞中升高,敲低其表达可抑制宫颈癌细胞侵袭和转移能力,这可能与其能调控AKT/mTOR信号通路活性有关。 Objective To investigate the expression of long noncoding RNA gastric cancer high expression transcript 1 (LncRNA GHET1) in cervical cancer and its effects on cervical cancer cell biological behavior.Methods Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) was used to detect the expression of GHET1 in tissues and cells,and the relationship between the expression of GHET1 and patient pathological data was analyzed.Hela cells were divided into si-NC group (transfected with negative control) and si-GHET1 group (transfected with si-GHET1).Transwell experiment was used to detect cell invasion ability,and scratch experiment was used to detect cell migration ability,and Western blot was used to detect protein content in the cells.Results Compared with the adjacent normal tissues,the expression of GHET1 was increased in cervical cancer tissues and cells (1.02±0.09 vs 2.96±0.26);compared with low expression of GHET1,high expression of GHET1 was related to high International Federation of Obstetrics and Gynecology(FIGO) grade (Ⅲ+Ⅳ),deep muscle layer infiltration,and lymph node metastasis.The expression level of GHET1in si-NC group and si-GHET1 group cells were 1.00±0.13 and 0.46±0.10,the number of cells passing through the membrane were 87.53±12.53 and 32.56±8.96;the scratch distances of the cells were (7.15±1.11) and(3.02±0.86)μm,and the relative expression levels of phospho-mammalian target of rapamycin (p-m TOR) were0.99±0.09 and 0.49±0.09,respectively.The differences in the aforementioned indicators between the two groups were statistically significant (all P0.05).Conclusion GHET1 is upregulated in cervical cancer tissues and cells,and knocking down its expression can inhibit the invasion and migration ability of cervical cancer cells,which may be related to the regulation of AKT/m TOR signaling pathway activity.
