虎杖苷对过敏性鼻炎小鼠炎症反应的作用及机制

Effect and mechanism of polydatin on inflammatory response in mice with allergic rhinitis

摘要 目的 探讨虎杖苷对过敏性鼻炎(AR)小鼠的保护作用及其作用机制。方法 将小鼠随机分为对照组、模型组、地塞米松组和低、中、高剂量组,每组10只小鼠。除对照组外其余小鼠均构建AR模型,地塞米松组小鼠每天灌胃1 mg·kg^(-1)的地塞米松,低、中、高剂量组小鼠分别每天灌胃25、50和100 mg·kg^(-1)的虎杖苷,对照组和模型组小鼠均灌胃给予等体积的磷酸盐缓冲液。在最后1次卵清蛋白鼻内刺激1 h后,记录5 min内打喷嚏和摩擦鼻腔的情况,以试剂盒法检测血清炎症细胞因子及免疫球蛋白E(IgE)表达水平,以蛋白质印迹法检测各组小鼠鼻黏膜相关蛋白表达情况,以活性氧(ROS)试剂盒检测ROS水平,试剂盒检测鼻黏膜组织氧化应激相关指标水平。结果 对照组、模型组、地塞米松组和高剂量组打喷嚏次数分别为(0.80±0.11)、(21.03±2.69)、(4.69±0.59)和(7.44±0.98)次,IgE水平分别为(140.22±15.23)、(367.49±30.29)、(168.13±14.99)和(177.34±16.79) pg·mL^(-1),白细胞介素6水平分别为(23.41±3.10)、(83.49±8.11)、(33.44±4.06)和(40.05±4.69) pg·mL^(-1),紧密连接蛋白(Occludin)蛋白水平分别为0.97±0.10、0.23±0.04、0.86±0.09和0.92±0.10,ROS水平分别为(0.93±0.04)、(3.19±0.30)、(1.24±0.14)和(1.29±0.13) RFU·mg^(-1),Nod样蛋白受体3(NLRP3)蛋白表达水平分别为0.34±0.05、0.94±0.08、0.40±0.05和0.42±0.04;以上指标,模型组与对照组比较,地塞米松组和高剂量组分别与模型组比较,差异均有统计学意义(均P<0.05)。结论 虎杖苷可能通过介导ROS/NLRP3/Caspase-1引起的细胞焦亡通路,抑制氧化应激、减轻AR炎症反应、改善鼻黏膜屏障功能。 Objective To investigate the protective effect of polydatin on allergic rhinitis (AR) mice and its mechanism.Methods Mice were randomly divided into control group,model group,dexamethasone(DEX) group,experimental-L,-M and-H groups,with 10 mice in each group.AR models were established for all the other mice except the control group.The DEX group were given 1 mg·kg^(-1)of dexamethasone per day;the experimental-L,-M and-H groups were given 25,50and 100 mg·kg^(-1)of polydatin per day,respectively;the control group and model group were given the same amount of phosphate buffer solution.After the last ovalbumin intranasal stimulation for 1 h,the number of sneezing and rubbing within 5 min were recorded.Serum inflammatory cytokines and immunoglobulin E (IgE) expression levels were detected by kit method;nasal mucous membrane related protein expression were detected by Western blot method;reactive oxygen species (ROS) level was detected by ROS kit.The kits were used to detect oxidative stress related indexes in nasal mucosa.Results The sneezing times of control group,model group,DEX group,experimental-H group were 0.80±0.11,21.03±2.69,4.69±0.59 and 7.44±0.98,respectively;IgE levels were (140.22±15.23),(367.49±30.29),(168.13±14.99) and (177.34±16.79) pg·m L^(-1),respectively;interleukin-6 levels were (23.41±3.10),(83.49±8.11),(33.44±4.06) and (40.05±4.69) pg·m L^(-1),respectively;the Occludin levels were0.97±0.10,0.23±0.04,0.86±0.09 and 0.92±0.10,respectively;ROS levels were (0.93±0.04),(3.19±0.30),(1.24±0.14) and (1.29±0.13) RFU·mg^(-1),respectively;the Nod-like protein receptor 3(NLRP3) protein expression levels were 0.34±0.05,0.94±0.08,0.40±0.05 and 0.42±0.04,respectively.The above indexes were compared between the model group and the control group,the differences were statistically significant (all P0.05).DEX group,experimental-L group,experimental-M group and experimental-H group were compared with model group,respectively,and the differences were statistically significant (all P0.05).Conclusion Polydatin may inhibit oxidative stress,reduce AR inflammation and improve nasal mucosal barrier function by mediating ROS/NLRP3/Caspase-1-induced pyroptosis pathway.
