摘要
目的 基于网络药理学分析余甘子治疗轻症急性胰腺炎作用机制,为临床的药物使用、动物实验、新药提取提供依据。方法 采用TCMSP(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform)数据库获取余甘子的主要活性成分及主要靶点;通过Gencards据库获取轻症急性胰腺炎的疾病靶点;利用Venny平台获取余甘子和轻症急性胰腺炎的共同靶点;在String数据库构建PPI蛋白数据网络;用Metascape平台采用GO与KEGG分析;用Cytoscape 3.7.2软件构建“靶点-通路”网络,分析重要通路;最终获取治疗轻症急性胰腺炎最重要的靶点。结果 余甘子治疗轻症急性胰腺炎的主要活性成分是鞣花酸、儿茶素、山柰酚、槲皮素;关键靶点有MAPK1、RELA、AKT1、JUN、TNF、IL6;主要生物过程有对无机物的反应、运动的正向调节、对激素的反应、对细菌源性分子的反应、对活性氧的反应;主要作用于蛋白激酶活性、蛋白质同源二聚化活性、磷酸转移酶活性,醇基为受体、DNA转录因子结合、蛋白质丝氨酸/苏氨酸/酪氨酸激酶活性等分子功能;主要富集的通路有癌症的途径、糖尿病并发症中的AGE-RAGE信号通路、PI3K-Akt信号通路、癌症中的蛋白多糖、肿瘤坏死因子信号通路、MAPK信号通路等。结论 通过网络药理学研究初步揭示了余甘子治疗轻症急性胰腺炎是多成分、多靶点、多通路共同作用的结果,为余甘子的药用提供了一定临床依据。
Objective To analyze the mechanism of Yuganzi in the treatment of mild acute pancreatitis based on network phar-macology,and to provide basis for clinical drug use,animal experiment and new drug extraction.Methods TCMSP(traditional Chinese medicine systems pharmacy database and analysis platform)database was used to obtain the main active components and main targets of Phyllanthus emblica;The disease targets of mild acute pancreatitis were obtained through gencards database;U-sing Venny platform to obtain the common targets of Phyllanthus emblica and mild acute pancreatitis;Construct PPI protein data network in string database;Go and KEGG analysis with metascape platform;Use Cytoscape 37.2 the software builds a"target pathway"network and analyzes important pathways;Finally,the most important target for the treatment of mild acute pancreatitis will be obtained.Results The main active components of Phyllanthus emblica in the treatment of mild acute pancreatitis were el-lagic acid,catechin,kaempferol and quercetin;The key targets are mapkl,rela,AKTI,Jun,TNF and IL6;The main biologi-cal processes include the reaction to inorganic substances,the positive regulation of movement,the reaction to hormones,the re-action to bacterial derived molecules and the reaction to reactive oxygen species;It mainly acts on protein kinase activity,protein homodimerization activity,phosphotransferase activity,alcohol group as receptor,DNA transcription factor binding,protein ser-ine/threonine/tyrosine kinase activity and other molecular functions;The major enrichment pathways include cancer pathway,ACE-RAGE signaling pathway in diabetes complications,PI3K-Akt signaling pathway,proteoglycan in cancer,tumor necro-sis factor signaling pathway and MAPK signaling pathway.Conclusion Through the network pharmacology research,it is prelimi-narily revealed that the treatment of mild acute pancreatitis by Phyllanthus emblica is the result of the joint action of multiple components,multiple targets and multiple pathways,which provides a certain clinical basis for the medicinal use of Phyllanthus emblica.
