犀角地黄汤对A群链球菌感染小鼠SOCS-1表达影响

Effect of Xijiao Dihuang Decoction(犀角地黄汤)on the Expression of SOCS-1 in GAS Infected Mice

摘要 目的观察细胞因子信号转导抑制因子1(SOCS-1)在小鼠A群链球菌(GAS)感染模型表达情况,分析犀角地黄汤对SOCS-1表达的影响。方法将36只小鼠随机分为3组(空白组、模型组、治疗组),每组12只,模型建立成功后,空白组和模型组均正常饲养,治疗组给予犀角地黄汤(0.4 mL/次,含生药1.5 g/mL,2次/d),分4个时点(造模后12、24、48、72 h)处死小鼠(每组3只/时点),每只小鼠采集感染部位皮肤软组织标本3份,运用实时定量PCR技术和蛋白印迹(Western blot)实验分别观察不同时间段SOCS-1 mRNA及蛋白的表达情况。在上述工作基础上,运用犀角地黄汤干预GAS感染的小鼠,每日早晚用药各1次,观察用药12、24、48、72 h后SOCS-1 mRNA及蛋白的表达情况,各组进行数据对比,分析犀角地黄汤对SOCS-1 mRNA及蛋白的表达影响。结果SOCS-1mRNA:与12、24、72 h对照组相比,模型组及治疗组SOCS-1 mRNA表达均明显升高,P<0.05;与24、72 h模型组相比,治疗组SOCS-1 mRNA表达下降明显,P<0.05。SOCS-1蛋白:与12、24、72 h对照组相比,模型组及治疗组SOCS-1蛋白表达水平均明显升高,P<0.05;与24、48、72 h模型组相比,治疗组SOCS-1蛋白表达水平下降明显,P<0.05。结论犀角地黄汤可以显著下调SOCS-1表达。 Objective To observe SOCS-1 expression,and analyze the effect of Xijiao Dihuang Decoction(犀角地黄汤)on SOCS-1 expression of GAS infection model in mice.Methods Thirty-six mice were randomly divided into three groups(blank,model,and treatment groups),with 12 mice in each group.After establishing the model,the blank and model groups were raised typically.The treatment group was given rhinoceros horn Rehmannia decoction(0.4 mL/time,containing 1.5 g/mL of crude drug,twice/day),and the mice were killed at four-time points(12,24,48,72 h after the establishment of the model)(3 mice/time point/group).Three skin and soft tissue samples of the infected site were collected from each mouse;pathological observation.SOCS-1 mRNA and protein expression were observed using quantitative PCR technology and Western blot experiment respectively.On the basis of the above work,the expression of SOCS-1 mRNA and protein was observed after 12,24,48 and 72 h.Data were compared in each group to analyze the effect of Xijiao Dihuang Decoction on SOCS-1 mRNA and protein expression.Results SOCS-1 mRNA:Compared with the control group,the SOCS-1 mRNA of the model group and the intervention group increased significantly at 12,24 and 72 h(P<0.05);The SOCS-1 mRNA of the intervention group was significantly lower than that of model group at 24 h and 72 h(P<0.05).SOCS-1 protein:Compared with the control group,the SOCS-1 protein of the model group and the intervention group increased significantly at 12,24 and 72 h(P<0.05);Compared with the intervention group and the model group at 24 h,48 h and 72 h,the SOCS-1 protein of the intervention group decreased significantly(P<0.05).Conclusion Xijiao Dihuang Decoction can significantly down regulate the expression of SOCS-1 in the early stage of GAS infectedmice.
