厚朴姜制前后化学成分UPLC-Q-TOF/MS分析 被引量:2

Analysis of Chemical Compositions in Houpo(Magnoliae Officinalis Cortex)Before and after Processingwith Gingerby UPLC-Q-TOF-MS

摘要 目的借助超高效液相色谱-四级杆-飞行时间串联质谱(UPLC-Q-TOF/MS)对厚朴及其姜制品的成分进行快速分类及鉴定,分析炮制前后的化学成分变化。方法采用Waters ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7μm),以0.1%甲酸水溶液(A)和0.1%甲酸乙腈(B)进行梯度洗脱,在正负离子模式下扫描,结合对照品、数据库及相关文献,运用MassLynx软件对厚朴及姜制品进行化学成分鉴定,分析主要成分裂解过程;并结合软件进行主成分分析和差异性分析,筛选得到厚朴炮制前后差异成分。结果鉴定出38种成分,并筛选出10种主要差异性成分,其中厚朴木脂素A、木兰醛B经姜制后不存在,厚朴木脂素E、和厚朴酚、厚朴酚、辣薄荷基厚朴酚含量减少,武当木兰碱、木兰箭毒碱、木兰花碱、降荷叶碱含量增加。结论厚朴中主要含有苷类、木脂素类及生物碱类成分,姜制后生物碱类和木脂素类成分产生了量变,这可能是导致厚朴姜制前后功效不同的主要原因,为厚朴临床应用提供了一定科学依据。 Objective The components of Houpo(Magnoliae Officinalis Cortex)and its ginger products were rapidly classified and identified by ultra-high performance liquid chromatography quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF/MS)to analyze the chemical composition changes before and after processing.Methods Gradient elution was performed on a Waters ACQUITY UPLC BEH Crs column(2.1 mm×100 mm,1.7μm)with 0.1%formic acid in water(A)and 0.1%formic acid in acetonitrile(B),and scanning was performed in positive and negative ion mode.MassLynx software was used to identify the chemical components of Houpo(Magnoliae Officinalis Cortex)and its ginger products to analyze the cracking process of main components.Then combined with the software,principal component analysis and difference analysis were carried out,and the difference components before and after processing of Houpo(Magnoliae Officinalis Cortex)were obtained by screening.Results A total of 38 components were identified,of which 10 were major differential components before and after processing.Magnoliain A and magnolialdehyde B do not exist after being processed by ginger,and the contents of magnolia E,honokiol,honokiol,and spicy menthyl honokiol are reduced,while the content of anthocyanine and norfolate increased.Conclusion Houpo(Magnoliae Officinalis Cortex)mainly contains glycosides,lignans and alkaloids,and the quantity of alkaloids and lignins changed after processing with ginger,which may be the main reason for the different effects of Houpo(Magnoliae Officinalis Cortex)before and after processing,and provide a certain scientific basis for the clinical application of Houpo(Magnoliae Officinalis Cortex).
