电针阳陵泉穴对缺血性脑卒中大鼠运动功能及血管新生相关因子的影响

Effect of Electroacupuncture at Yanglingquan(GB34)on Motor Function and Expression of Angiogenesis Related Factors in Rats with Ischemic Stroke

摘要 目的观察电针阳陵泉穴对缺血性脑卒中大鼠运动功能及血管新生相关因子血管内皮生长因子(vascular endothelial growth factor,VEGF)、血管生成素-1(angiogenin-1,Ang-1)表达的影响,并探讨电针的作用机制。方法将雄性SD大鼠随机分为假手术组、模型组、电针组,采用改良线栓法制备大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)大鼠模型,其中假手术组与模型组无治疗,电针组大鼠给予电针阳陵泉穴干预14 d,各组基于造模术后1、7、14 d分为3个亚组,每个亚组各8只,观察各组在不同时点神经功能缺损评分及运动评分的变化,应用HE染色观察各组大鼠脑组织形态学变化,通过ELISA法、RT-PCR法分别检测各组大鼠血清、脑组织VEGF、Ang-1表达水平。结果与模型组比较,治疗7、14 d电针组大鼠神经功能缺损评分、平衡木以及网屏评分明显降低(P<0.05),治疗14 d转棒评分也明显降低(P<0.01);电针组于治疗7、14 d能明显上调MCAO大鼠血清Ang-1及脑组织Ang-1 mRNA的表达(P<0.05),且在不同时点电针组能显著升高脑组织VEGF mRNA的表达(P<0.05),而对血清VEGF的上调效应在治疗14 d最为明显(P<0.05)。结论电针阳陵泉穴可改善缺血性脑卒中大鼠神经功能缺损及运动功能障碍,其机制可能与调控血管新生相关因子的表达有关。 Objective To explore the mechanism of electroacupuncture by observing the effect of electroacupuncture at Yanglingquan(GB34)on motor function and the expression of angiogenesisrelated factors such as VEGF and Ang-1 in rats with ischemic stroke.Methods Male SD rats were randomly divided into sham operation group,model group and electroacupuncture(EA)group,and the rat model of middle cerebral artery occlusion(MCAO)was prepared by modified suture occlusion method.Among of three groups,the EA group was treated by electroacupuncturing at bilateral Yanglingquan(GB34)for 14 days while the sham operation group and the model group without treatment.Each group was divided into 3 subgroups with 8 rats in each subgroup based on 1 day,7 days and 14 days after modeling.The changes of neurological deficit scores and motor scores at different time points in each group were observed.By the method of HE staining,the morphological changes of brain tissue in each group were observed.The expression levels of VECF and Ang-1 in serum and brain tissue were tested by the method of ELISA and RT-PCR.Results Compared with the model group,the neurological deficit scores,balance beam scores and screen scores of rats in EA group were significantly decreased after 7 and 14 days of treatment(P<0.05),the rod turning scores were also significantly decreased(P<0.01)after 14 days of treatment.The EA group could significantly up-regulate the expression of Ang-1 mRNA in serum and brain tissue of MCAO rats after 7 and 14 days of treatment(P<0.05),and increase the expression of VECF mRNA in brain tissue at different time points(P<0.05).However,the upregulation effect on serum VECF was most obvious after 14 days of treatment(P<0.05).Conclusion Electroacupuncture at Yanglingquan(GB34)can improve neurological deficit and motor dysfunction in rats with ischemic stroke,and its mechanism may be related to regulating the expression of angiogenesis related factors.
