辛夷影响变应性鼻炎Th1/Th2细胞因子实验研究 被引量:2

The influence of Xinyi on Th1/Th2 cytokines in mice with allergic rhinitis

摘要 目的:探讨辛夷对变应性鼻炎(Allergic Rhinitis,AR)1型辅助性T细胞(T helper type 1 cell,Th1)和2型辅助性T细胞(T helper type 2 cell,Th2)细胞因子的作用及机制。方法:将BALB/c小鼠分为正常对照组、AR组和辛夷干预AR组,每组8只。AR小鼠应用辛夷精油经鼻腔滴药,末次激发结束后30 min内观察小鼠行为学改变,记录打喷嚏、抓鼻挠痒次数和鼻腔分泌物等鼻部症状并评分。末次激发24小时后收集血标本,采用酶联免疫吸附法(Enzyme Linked Immunosorbent Assay,ELISA)检测外周血白细胞介素-2(Interleukin-2,IL-2)、干扰素-γ(Interferon-γ,IFN-γ)、白细胞介素-4(Interleukin-4,IL-4)、白细胞介素-5(Interleukin-5,IL-5)的浓度。结果:AR小鼠打喷嚏、抓鼻挠痒和流鼻涕等鼻部症状明显,经辛夷干预后鼻部症状减轻,AR组与正常对照组比较、辛夷干预AR组与AR组比较差异均有统计学意义(P<0.05)。AR小鼠外周血IL-4、IL-5浓度升高,IL-2、IFN-γ浓度降低;经辛夷干预,IL-4、IL-5浓度下降,IL-2、IFN-γ浓度升高,AR组与正常对照组比较、辛夷干预AR组与AR组比较差异均有统计学意义(P<0.05)。结论:辛夷可降低Th2细胞因子浓度,升高Th1细胞因子浓度,从而改善Th1/Th2细胞平衡,减轻AR炎症反应。 Objective:To explore the mechanism of Xinyi(Flos Magnoliae)on Th1/Th2 cytokines in mice with allergic rhinitis.Methods:BALB/c mice were divided into the normal control group,AR group and Xinyi intervention AR group with 8 mice in each group.The behavioral changes of the mice were observed within 30 minutes after the end of the last stimulation,and theO symptoms such as sneezing,scratching and nasal secretions were recorded.The ELISA Method was used to determine the concentration of IL-2,IFN-γ,IL-4 and IL-5 in the peripheral blood of mice with allergic rhinitis who were treated by the volatile oil of Xinyi.Results:The number of sneezing,scratching and nasal secretions was decreased after treated by the volatile oil of Xinyi.The differences between the AR group and the the normal control group,the Xinyi intervention AR group and the AR group were statistically significant(P<0.05).The expressions of IL-4 and IL-5 in the peripheral blood of mice with allergic rhinitis were decreased,and the expressions of IL-2 and IFN-γwere increased after treated with the volatile oil of Xinyi.The differences between the AR group and the the normal control group,the Xinyi intervention AR group and the AR group were statistically significant(P<0.05).Conclusion:The expression of Th2 cytokines was decreased and the expression of Th1 cytokines was increased after treated,thus regulating the balance of Th1/Th2 cells,and reducing the inflammation of allergic rhinitis.
