基于网络药理学探讨复方蛇龙胶囊治疗膜性肾病的作用机制

摘要 目的:利用网络药理学探究复方蛇龙胶囊治疗膜性肾病的作用机制。方法:基于中药系统药理学数据库与分析平台(TCMSP)获取复方蛇龙胶囊中三味组成药材白花蛇舌草、穿山龙、鬼箭羽的靶基因,通过基因组注释数据库平台(GeneCards)和OMIM数据库收集膜性肾病的靶基因。将复方蛇龙胶囊和膜性肾病的靶基因在Venny 2.1中取交集,通过STRING和Cytoscape进行蛋白质-蛋白质相互作用网络构建及关键共有靶点筛选,并通过Metascape数据库对共同靶点进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)富集分析。结果:本研究共获得符合标准有效活性成分5个,复方蛇龙胶囊-膜性肾病交集靶点84个,通过Cytoscape拓扑分析得到核心靶点18个,主要富集于生物过程、细胞组成、分子功能及164条信号通路上,主要表现在癌症通路、糖尿病并发症中的晚期糖基化终末产物-糖基化终末产物受体(AGE-RAGE)信号通路、丝裂原活化蛋白激酶(MAPK)信号通路、缺氧诱导因子-1α(Hypoxia Inducible Factor-1 Subunit Alpha,HIF-1α)信号通路、核因子-κB(Nuclear Factor kappa-B,NF-κB)信号通路、化学致癌作用-受体激活。结论:复方蛇龙胶囊可能通过5个主要活性成分作用于18个核心靶点影响生物过程,调节磷脂酰肌醇3激酶-蛋白激酶B(PI3K-Akt)信号通路、p38MAPK信号通路、HIF-1信号通路、NF-κB信号通路来发挥治疗膜性肾病的作用。 Objective:To explore the mechanism of the Fufang Shelong capsules(复方蛇龙胶囊)in the treatment of membranous nephropathy by network pharmacology.Methods:The target genes of Baihua Sheshe Cao(Herba Hedyoti Diffusae),Chuanshanlong(Rhizoma Dioscoreae Nipponicae),Guijianyu(Ramulus Euonymi)in the Fufang Shelong capsules were obtained based on the TCM medicine System Pharmacology Platform(TCMSP),and the target genes of membranous nephropathy were collected through the Genome Annotation database platform(GeneCards)and OMIM database.The target genes of the Fufang Shelong capsules and membranous nephropathy were intersected by Venny 2.1,and the protein-protein interaction(PPI)network was constructed by STRINGOandCytoscape,and the key common targets were screened.GO enrichment analysis and KEGG enrichment analysis were performed on the common targets by Metascape database.Results:In this study,a total of 5 active ingredients met the standard,84 intersection targets of the Fufang Shelong capsules and membranous nephropathy were obtained,and 18 core targets were obtained by Cytoscape topological analysis,which were mainly enriched in biological processes,cell composition,molecular function and 164 signaling pathways.It is mainly manifested in cancer pathway,AGE-RAGE signaling pathway in diabetic complications,MAPK signaling pathway,HIF-1 signaling pathway,NF-κB signaling pathway,chemical carcinogenation-receptor activation.Conclusion:The Fufang Shelong capsules may play a role in the treatment of membranous nephropathy by regulating PI3K-Akt signaling pathway,p38MAPK signaling pathway,HIF-1 signaling pathway and NF-κB signaling pathway through the action of 5 main active components on 18 core targets.
