摘要
目的探讨STAT3和自噬相关蛋白Beclin1、p62、LC3B在结肠癌组织中的表达情况,并分析它们之间的相关性。方法免疫组化方法检测结肠癌组织(n=86)、癌旁组织(n=86)及正常结肠组织(n=86)包埋蜡块中STAT3、Beclin1、p62、LC3B的阳性表达率,并分析STAT3与自噬指标在结肠癌中表达的相关性。结果相比于癌旁及正常组织,在结肠癌组织中,STAT3、p62、LC3B的表达水平明显升高,Beclin1的表达水平降低,且STAT3与p62、LC3B表达水平呈正相关,与Beclin1表达水平呈负相关。结论在结肠癌中,STAT3与自噬蛋白之间存在可调节的关系,通过这种联系促进或抑制结肠癌的发生及发展,干预STAT3-自噬通路可能为疾病治疗提供新的思路。
Objective To investigate the expression of STAT3 and autophagy related proteins Beclin1,p62,LC3B in colon cancer tissue,and analyze their correlation.Methods Immunohistochemical methods was used to detect the positive expression rates of STAT3,Beclin1,p62 and LC3B in colon cancer tissue(n=86)paracancerous tissue(n=86)and normal colon tissue(n=86)embedded wax blocks.The correlation between STAT3 and autophagy was analyzed in colon cancer.Results Compared with normal tissues and paracancerous tissues,the expression levels of STAT3,p62 and LC3B were significantly increased in colon cancer tissues,while the expression level of Beclin1 was reduced.STAT3 was positively correlated with the expression levels of p62,LC3B,and negatively correlated with the expression level of Beclin1.Conclusion In colon cancer,there is an adjustable relationship between STAT3 and autophagy protein,through which the occurrence and development of colon cancer could be promoted or inhibited.Therefore,the intervention in STAT3-autophagy pathway could be a new approach to the treatment of colon cancer.
