参麦注射液对小分子抗血管生成靶向药增效、减毒的药效学分析

摘要 目的观察参麦注射液对小分子抗血管生成靶向药DC-101的减毒、增效作用。方法将皮下接种CT-26的小鼠40只随机分为四组,每组各10只,分别为对照组、DC-101组、参麦注射液组、联合治疗组,分别给予腹腔注射0.9%氯化钠溶液、DC-101、参麦注射液、DC-101+参麦注射液治疗。给药后的第3、7、10、14天测定肿瘤体积;给药后14 d处死小鼠,测定瘤重、脾脏指数、外周血白细胞数量以及肿瘤组织血管密度和VEGF表达。结果给药14 d后,DC-101组、联合用药组瘤重显著低于对照组(P<0.05),联合用药组瘤重略低于DC-101组(P>0.05);联合用药组脾脏指数与外周白细胞计数显著高于DC-101组(P<0.05);联合治疗组的微血管密度和VEGF表达评分显著低于对照组(P<0.05),略低于DC-101组(P>0.05)。结论在结肠癌小鼠模型中,参麦注射液能增加DC-101对肿瘤生长的抑制作用,有效改善DC-101所导致的白细胞下降,抑制肿瘤血管新生。 Objective To observe the toxicity reducing and efficacy enhancing of Shenmai Injection combined with small molecule antiangiogenesis targeted drug DC-101.Methods Forty mice subcutaneously inoculated with CT-26 were randomly divided into four groups,10 mice in each group,the control group,the DC-101 group,the Shenmai injection group and the combined treatment group,these four groups were separately treated with normal saline,DC-101,Shenmai injection and DC-101+Shenmai injection.Then the tumor volume was measured on the 3rd,7th,10th and 14th days after administration,and the mice were killed on the 14th day after administration to determine the tumor weight,spleen index and the number of peripheral blood leukocytes,and the vascular density and VEGF expression of tumor tissue.Results After 14 days of administration,the tumor weight of the DC-101 group and the combined treatment group was significantly lower than that of the control group(P<0.05),and that of the combined treatment group was slightly lower than that of the DC-101 group(P>0.05).The spleen index and peripheral white blood cell count in the combined treatment group were significantly higher than those in the DC-101 group(P<0.05).The vascular density and VEGF expression of the combined treatment group were significantly lower than those of the control group(P<0.05),and little lower than the DC-101 group(P>0.05).Conclusion In colon cancer mice model,Shenmai injection can increase the inhibitory effect of DC-101 on tumor growth,and effectively improve the leukopenia induced by DC-101,and inhibit the tumor angiogenesis.
