摘要
目的:观察水族药南烛根对骨折大鼠的血管生成作用,探讨其对骨折的治疗作用。方法:采用锯开法建立大鼠胫骨干骨折模型。将大鼠随机分成假手术组、模型对照组,阳性对照组(接骨七厘片20 mg/kg)、南烛根水提液高、中、低剂量组(400 mg/kg、200 mg/kg、100 mg/kg),每组10只。建模后第2天起,各给药组按照相应剂量配置溶液灌胃,每天2次。假手术组和模型对照组不给予药物,正常饮食。连续给药2周。观察大鼠骨痂镜下形态和测量血管面积比,免疫组化法检测骨痂组织VEGF、VEGFR-2阳性表达量。结果:与假手术组比较,模型对照组、阳性对照组、南烛根水提液低剂量组、中剂量组、高剂量组大鼠骨痂血管面积百分比、骨痂VEGF、VEGFR-2阳性表达显著增高(P<0.05);与模型对照组比较,阳性对照组、南烛根水提液高剂量组大鼠骨痂血管面积百分比、骨痂组织VEGF、VEGFR-2阳性表达显著增高(P<0.05);与阳性对照组、南烛根水提液高剂量组比较,南烛根水提液低剂量组、中剂量组骨痂血管面积百分比、骨痂组织VEGF阳性表达明显降低(P<0.05)。与阳性对照组比较,南烛根水提液低剂量组、中剂量组、高剂量组骨痂VEGFR-2阳性表达明显降低(P<0.05)。结论:南烛根水提液可明显促进骨痂组织VEGF、VEGFR-2阳性表达,内皮细胞分裂,促进血管新生,从而达到促进骨折愈合的作用。
Objective This study aimed to observe the therapeutic effect of shui medicine Vaccinium bracteatum Thunb for promoting angiogenesis in tibia fracture rats.Methods The model of tibial shaft fracture will be established by sawing.60 rats were randomly divided into sham operation group,model control group,positive control group(Jiegu Qili Tablet 20 mg/kg),and high,medium,low dose groups of water extract of Vaccinium bracteatum Thunb.(400 mg/kg、200 mg/kg、100 mg/kg),with 10 in each.Except for the sham operation group,all groups will be gavaged according to the corresponding dose,1time/d,continuous for 2 weeks.After the treatment,Observe the morphology of rat callus under microscope and measure the Morphology of bone callus and the percentage of vascular area were measured.The relative expression of VEGF and VEGFR-2 protein was detected by immunohistochemistry.Results Compared with the sham operation group,the percentage of vascular area and the positive expression of VEGF and VEGFR-2 in model control group,positive control group,low dose group,medium dose group and high dose group were significantly higher(P<0.05).Compared with the model control group,the percentage of vascular area and the positive expression of VEGF and VEGFR-2 in positive control group,low dose group,medium dose group and high dose group were significantly higher(P<0.05).Compared with the positive control group and the high dose group,the percentage of vascular area and Vaccinium bracteatum the positive expression of VEGF in low dose group,medium dose group were significantly lower(P<0.05).Compared with the positive control group,the positive expression of VEGFR-2 in low dose group,medium dose group and high dose group were significantly lower(P<0.05).Conclusions Extraction ofshui medicine Vaccinium bracteatum Thunb.could significantly enhance the positive expression of VEGF and VEGFR-2 in bone tissue,also could promote endothelial cell division and angiogenesis,which mechanism is advantage for fracture healing.
