合肥市蜱系统发育分析及安徽省一蜱种新纪录

Phylogenetic analysis of ticks in Hefei City and a new tick species record in Anhui Province

目的了解安徽省合肥市蜱类系统发育情况,并对安徽省一新纪录种进行描述。方法选取合肥市长丰县、蜀山区、肥西县作为采样点,采集蜱样进行形态学鉴定后,选取部分样本,用PCR方法扩增线粒体核糖体核糖核酸大亚基(16S rRNA)和细胞色素C氧化酶I(COI)基因,测序后进行相似性及系统发育分析,并对发现的安徽省硬蜱科一新纪录种进行鉴定。结果长丰县、蜀山区、肥西县3个采样点共采集到18只蜱样,其中长丰县1只,蜀山区11只,肥西县6只。形态学鉴定为褐黄血蜱12只,长角血蜱5只,豪猪血蜱1只。16S rRNA和COI基因序列与Genbank上下载的序列比对显示,本次所获得长角血蜱、褐黄血蜱、豪猪血蜱分别聚于不同分支,长角血蜱与豪猪血蜱亲缘关系较近,与褐黄血蜱亲缘关系较远。采集到的豪猪血蜱为安徽新纪录种。结论初步了解了合肥市蜱类构成,安徽省蜱类新增纪录种为豪猪血蜱。

Objective To understand the phylogenetic of ticks in Hefei City,and identify a tick species newly recorded in Anhui Province.Methods Ticks were collected from Changfeng County,Shushan District and Feixi County in Hefei.Morphological identification was performed on the collected ticks.The large subunit of mitochondrial ribosomal RNA(16S rRNA)and cytochrome C oxidase I(COI)genes were amplified by PCR and sequenced for the phylogenetic analysis.A tick species was newly recorded in Anhui Province.Results A total of 18 ticks were collected in Hefei,one from Changfeng County,11 ticks from Shushan District and 6 ticks from Feixi County.Morphological identification told 12 Haemaphysalis flava,5 Haemaphysalis longicornis,and 1 Haemaphysalis hystricis.The 16S rRNA and COI gene analysis revealed that Haemaphysalis longicornis,Haemaphysalis flava and Haemaphysalis hystricis clustered in different branches.Haemaphysalis longicornis was closely related to Haemaphysalis hystricis,but distantly related to Haemaphysalis flava.Haemaphysalis hystricis was a newly recorded species in Anhui Province.Conclusion The tick composition in Hefei City has been initially assessed,and the newly recorded tick species in Anhui Province was Haemaphysalis hystricis.
