东海海参多糖酶解提取工艺优化及其抗氧化活性

摘要 以东海海参(Acaudina leucoprocta)干品为原料,探究东海海参多糖的最佳提取工艺,并评价其基本组成及抗氧化活性。利用单因素试验考察木瓜蛋白酶的酶解温度、酶解时间、酶添加量、料液比对东海海参多糖提取率的影响,利用正交试验设计对工艺参数进行优化,利用红外光谱对得到的东海海参多糖进行鉴定,并检测东海海参多糖对DPPH·和·OH的清除率,考察东海海参多糖的抗氧化性。结果表明,各因素对多糖提取率的影响大小顺序为酶添加量>酶解时间>酶解温度,优化的最佳提取工艺为料液比1∶20(g/mL)、酶解温度55℃、酶解时间5 h、酶添加量1.0%,此条件下,东海海参多糖提取率为(11.85±0.27)%;东海海参多糖对DPPH·和·OH的清除效应随其浓度增加而提高。结果表明,东海海参多糖具有较强的抗氧化活性。 Polysaccharides were extracted from dried Acaudina leucoprocta,and the basic composition and antioxidant activity of the extracted polysaccharides were studied.The effects of enzymolysis temperature and time,enzyme(papain)concentration,and solid-to-liquid ratio on the yield of A.leucoprocta polysaccharides were investigated by single factor tests,and the extraction conditions were optimized by orthogonal design.The obtained polysaccharides were identified by infrared spectroscopy,and the scavenging rate on DPPH·and·OH free radicals were determined to evaluate the antioxidant activity of the polysaccharides.The results showed that the effects of different factors on the yield of polysaccharides followed the trend of enzyme concentration>enzymatic hydrolysis time>enzymatic hydrolysis temperature.The extraction conditions were optimized as hydrolysis with 1.0%enzyme and solid-to-liquid ratio of 1∶20(g/mL)at 55°C for 5 h.Under these conditions,the yield of A.leucoprocta polysaccharides was(11.85±0.27)%.The scavenging effects of A.leucoprocta polysaccharides on DPPH·and·OH free radicals increased in a concentration-dependent manner.The results indicated that A.leucoprocta polysaccharides had strong antioxidant activity.
