摘要
目的:基于代谢组学方法观察痹宁汤对痛风性肾病小鼠肾脏代谢组学的影响,寻找潜在生物标志物及相关代谢通路,探讨痹宁汤改善痛风性肾病的作用机制。方法:基于LC-MS非靶向代谢组学技术,采用主成分(PCA)和正交偏最小二乘—判别分析法(OPLS-DA)分析空白组、模型组、痹宁汤组小鼠肾脏代谢物及其所参与的代谢通路,探讨可作为诊断痛风性肾病的潜在生物标志物及其代谢通路。结果:筛查得到了24个可作为诊断痛风性肾病的生物标志物,以及4条相关代谢通路。结论:痹宁汤可以通过对回调多种代谢产物的含量及调控甘油磷脂代谢、癌症中的胆碱代谢、逆行内源性大麻素信号、嘌呤代谢等代谢途径改善痛风性肾病。
Objective:To observe the effect of Bining Decoction(BD)on metabolites of kidney tissues in mice with gouty nephropathy based on LC-MS,explore potential biomarkers and related metabolic pathways,and search for the mechanism of BD in improving gouty nephropathy.Methods:Based on LC-MS,non targeted metabolomics technology,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS DA)were used to analyze the renal metabolites and metabolic pathways involved in the mice of blank group,model group,and BD group,exploring potential biomarkers and metabolic pathways that can be used as diagnostic markers for gouty nephropathy.Results:We obtained 24 biomarkers that can be used to diagnose gouty nephropathy and 4 related metabolic pathways by the screening.Conclusion:Bining decoction can improve gouty nephropathy by regulating the content of various metabolites and regulating glycerophospholipid metabolism,choline metabolism in cancer,retrograde endocannabinoid signaling,and purine metabolism.
