高效液相色谱-二极管阵列检测器法对不同区域洋葱中烟酸的质量评价

摘要 建立了高效液相色谱-二极管阵列检测器法测定洋葱中烟酸的方法,并对洋葱中不同区域的烟酸进行研究分析。样品用0.1%(体积分数,下同)甲酸水超声提取后,用Agilent ZORBAX Eclipse Plus C18液相色谱柱(250 mm×4.6 mm,5μm),以甲醇和0.1%甲酸溶液为流动相,流速为1.0 mL/min,柱温为35℃,进样量为10μL,二级管阵列检测器检测范围190~400 nm,检测波长为260 nm。烟酸在1~100μg/mL线性相关系数R为0.99991~0.99998,方法检出限为0.15 mg/kg,定量限为0.50 mg/kg,10、20、30μg/mL 3个浓度加标水平的回收率为98%~103%,相对标准偏差为0.08%~0.11%,提取和分析效果是报道文献方法的1.5~2倍。该方法提取方法简单、响应好、重现性和回收率好,具有优异的精密度、准确度,可用于洋葱中烟酸分析检测,对全国8大主产区洋葱中烟酸的分析研究发现甘肃酒泉产的含量最高。 A method for the determination of nicotinic acid in onion by high performance liquid chromatography with diode array detector was established,and nicotinic acid in onion from different regions was studied.After ultrasonic extraction of the sample with 0.1%formic acid water,Agilent ZORBAX Eclipse Plus C18 liquid chromatographic column(250 mm×4.6 mm,5μm)was used,with methanol and 0.1%formic acid solution as mobile phase,a flow rate of 1.0 mL/min,column temperature of 35℃,injection volume of 10μL.The detection range of the diode array detector was 190-400 nm,and the detection wavelength was 260 nm.The linear correlation coefficient R in 1-100μg/mL of the quercetin concentration was 0.99991-0.99998 and the detection limit of quercetin was 0.15 mg/kg,and the quantification limit of quercetin were 0.50 mg/kg.The recovery rate of the three concentration spiking levels of 10,20,and 30μg/mL was 98%-103%and the relative standard deviations were 0.08%-0.11%.The effect of extraction and analysis was 1.5-2 times that of the reported methods.The method had the advantages of simple extraction,good response,good reproducibility and recovery rate,excellent precision and accuracy,and could be used for the analysis and detection of niacin in onions.It could be used for the analysis and detection of nicotinic acid in onions.The analysis of nicotinic acid in onion from 8 major production areas in China found that the content of quercetin in Jiuquan was the highest.
