理中汤通过调控线粒体自噬改善非酒精性脂肪性肝病的作用机制研究

摘要 目的:旨在探讨理中汤对非酒精性脂肪性肝病(NAFLD)的治疗效果及其作用机制。方法:使用高脂饲料诱导NAFLD大鼠,收集其血液和肝脏样本,比较各组大鼠体重、肝脏湿重,并计算肝指数;观察肝脏病理学变化;检测血清转氨酶和血脂水平;通过Western Bolt检测大鼠肝脏中PINK1、Parkin、LC-3Ⅱ的蛋白表达水平。结果:与空白组相比,NAFLD大鼠体重、肝脏湿重、肝指数、转氨酶和血脂水平明显升高,PINK1、Parkin、LC-3Ⅱ蛋白的表达水平明显下降(P<0.01)。经理中汤干预后,各中药组的大鼠体重、肝脏湿重、肝指数和血清转氨酶、血脂水平均下降,PINK1、Parkin、LC-3Ⅱ蛋白的表达水平均回升(P<0.05),其中高剂量组效果最为显著(P<0.01)。肝组织病理学结果显示,模型组大鼠肝脏组织显示肝小叶结构被破坏,肝细胞肿胀,排列紊乱,胞浆内含有大量大小不等的脂肪空泡;而在不同剂量的中药治疗组中,大鼠肝组织病理学表现均有所改善,其中高剂量组效果最显著。结论:理中汤可通过调控线粒体自噬改善NAFLD疾病。 Objective:To investigate the therapeutic efficacy and underlying mechanisms of LiZhong Tang in the context of non⁃alcoholic fatty liver disease(NAFLD).Methods:High-fat feed was used to induce NAFLD in rats,Blood and liver sam⁃ples were collected to facilitate a comparative analysis of rat body mass and liver wet weight and calculate the liver index.Liver pa⁃thology was observed,while serum transaminase and blood lipid levels were measured.The protein expression levels of PINK1,Parkin,and LC⁃3Ⅱin rat liver were detected using Western Blot analysis.Results:Compared with the control group,the NAFLD rats exhibited a significant increase in body weight,liver wet weight,liver index,transaminase levels,and blood lipid levels.The expression levels of PINK1,Parkin,and LC3⁃II protein were significantly decreased(P<0.01).Following interven⁃tion with Lizhong Tang,rats in each herbal treatment group displayed a decrease in body weight,liver wet weight,liver index,se⁃rum transaminase,and blood lipid levels.The expression levels of PINK1,Parkin,and LC⁃3Ⅱrebounded(P<0.05),with the high⁃dose group demonstrating the most pronounced effects(P<0.01).Histopathological examination of liver tissue revealed that rats in the model group displayed disrupted hepatic lobule structure,swollen hepatocytes,disordered arrangement,and a multi⁃tude of varying⁃sized lipid vacuoles within the cytoplasm.Conversely,rats treated with different doses of the herbal remedy exhibit⁃ed improvements in liver tissue pathology,with the high⁃dose group showing the most notable enhancement.Conclusion:Lizhong Tang can improve NAFLD disease by regulating mitochondrial autophagy.
