LINC01308在卵巢癌中的表达及临床意义

摘要 目的研究LINC01308对卵巢癌细胞的增殖、侵袭、迁移等生物学行为的影响。方法通过实时聚合酶链反应(qRT-PCR)检测LINC01308在卵巢癌细胞系、卵巢癌组织及相应的正常组织中的表达。利用慢病毒载体介导的shRNA沉默LINC01308后,通过细胞增殖实验(CCK-8)、细胞迁移和侵袭实验(Trans-well)、划痕愈合实验来验证LINC01308对卵巢癌细胞增殖、迁移和侵袭能力的影响。建构裸鼠成瘤模型,研究体内LINC01308对卵巢癌成瘤能力的影响。结果LINC01308在卵巢癌组织中的表达水平明显高于癌旁正常组织的表达水平。卵巢癌细胞中LINC01308的表达水平均显著高于正常卵巢上皮细胞系。LINC01308表达水平越高,卵巢癌患者的预后越差。卵巢癌患者预后不良和LINC01308表达高水平相关。降低LINC01308的水平表达后,在CCK-8增殖实验的活性曲线中显示卵巢癌细胞的生长受到抑制;划痕愈合实验中显示卵巢癌细胞创面闭合率显著降低;在Transwell侵袭实验发现迁移至小室和黏附到膜底层的卵巢癌的数量明显减少,侵袭细胞数下降得更低;裸鼠成瘤中注射沉默LINC01308的卵巢癌细胞组的小鼠体内形成的瘤体体积及重量均明显小于对照组。结论LINC01308在卵巢癌组织中和卵巢细胞中的表达水平均显著升高。LINC01308与卵巢癌患者预后不良相关,LINC01308高表达水平与卵巢癌患者的总生存期呈负相关。LINC01308能促进卵巢癌细胞的增殖、迁移、侵袭等生物学能力,促进裸鼠体内卵巢癌的生长。 Objective To explore the effects of LINC01308 on the proliferation,invasion and migration of ovarian cancer cells.Methods The expression of LINC01308 in ovarian cancer cell lines,ovarian cancer tissues and corresponding normal tissues was detected by real-time polymerase chain reaction(RT-PCR).After silencing LINC01308 with lentiviral vector mediated shRNA,and the influence of LINC01308 on the proliferation,migration and invasion of ovarian cancer cells were verified by CCK-8 experiment,Transwell experiment and scratch healing experiment.The effect of LINC01308 on the tumorigenic ability of ovarian cancer in vivo was studied by constructing the tumorigenic model of nude mice.Results The expression level of LINC01308 in ovarian cancer was significantly higher than that in adjacent normal tissues.The expression level of LINC01308 in ovarian cancer cells was significantly higher than that in normal human ovarian epithelial cell lines.The higher the expression level of LINC01308,the worse the prognosis of patients with ovarian cancer.The poor prognosis of patients with ovarian cancer was related to the high level of LINC01308 expression.After silencing the expression of LINC01308,the activity curve of the CCK-8 experiment showed that the growth of ovarian cancer cells was inhibited.The wound healing experiment showed that the wound closure rate of ovarian cancer cells decreased significantly.The Transwell invasion experiment showed that the number of ovaries migrating to the chamber and adhering to the bottom of the membrane decreased significantly,and the number of invasive cells decreased even lower.The tumor volume and weight of nude mice injected with ovarian cancer cells with silenced LINC01308 were significantly smaller than those in the control group.Conclusion The expression level of LINC01308 in ovarian cancer and ovarian cells increased significantly.LINC01308 is associated with poor prognosis of patients with ovarian cancer.The high expression level of LINC01308 is negatively correlated with the overall survival of patients with ovarian cancer.LINC01308 could promote the proliferation,migration and invasion of ovarian cancer cells,and then promote the growth of ovarian cancer in nude mice.
