摘要
目的探讨复方芙蓉花叶提取物对烧伤后创面感染大鼠创面愈合的影响及其作用机制。方法选取45只大鼠,10只作为空白组,其余35只建立烧伤后创面感染模型,35只建模大鼠最终有30只建模成功,将最终建模成功的30只大鼠分为模型组、药物对照组、实验组,每组各10只。药物对照组给予庆大霉素,实验组给予4 g/L复方芙蓉花叶提取物,空白组、模型组给予普通无菌纱布,连续干预10 d后观察大鼠变化。结果与空白组比较,模型组、药物对照组、实验组创面愈合率、基质金属蛋白酶-2(MMP-2)、MMP-9、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、IL-6水平上升,血管内皮生长因子(VEGF)、Wnt1、β-连环蛋白(β-catenin)mRNA表达量及相对表达量下降(P<0.05);与模型组相比,药物对照组、实验组创面愈合率、VEGF水平、Wnt1、β-catenin mRNA表达量及相对表达量上升,MMP-2、MMP-9、TNF-α、IL-1β、IL-6下降(P<0.05);与药物对照组比较,实验组创面愈合率、VEGF水平、Wnt1、β-catenin mRNA表达量及相对表达量上升,MMP-2、MMP-9、TNF-α、IL-1β、IL-6下降(P<0.05)。结论烧伤后创面感染大鼠经复方芙蓉花叶提取物干预后创面愈合率上升,血管相关因子水平得到调节,炎症反应减轻,其机制可能与Wnt/β-catenin信号通路激活有关。
OBJECTIVE To investigate the effect of compound hibiscus flower leaf extract on wound healing in rats with post-burn trauma infection and its mechanism of action.METHODS Totally 45 rats were selected,10 rats were used as blank group,and the remaining 35 rats were used to establish a post-burn trauma infection model.Of the 35 modeled rats,30 rats were finally successfully modeled,which were divided into the model group,the drug control group and the experimental group,with 10 rats in each group.The drug control group was given gentamicin,the experimental group was given 4 g/L compound hibiscus flower leaf extract,the blank group and model group were given ordinary sterile gauze,and the rats were observed after continuous intervention for 10 days.RESULTS Compared with the blank group,the wound healing rate,matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)levels increased in the model group,drug control group and experimental group,while vascular endothelial growth factor(VEGF)levels,Wnt1,β-catenin(β-catenin)mRNA expression and relative expression decreased(P<0.05).Compared with the model group,the wound healing rate,VEGF level,Wnt1,β-catenin mRNA expression and relative expression in the drug control group and experimental group increased,while MMP-2,MMP-9,TNF-α,IL-1β,IL-6 level decreased(P<0.05).Compared with the drug control group,the wound healing rate,VEGF level,Wnt1,β-catenin mRNA expression and relative expression in the experimental group increased,while MMP-2,MMP-9,TNF-α,IL-1β,IL-6 levels in the experimental group decreased(P<0.05).CONCLUSION The rate of wound healing was increased,the levels of vascular-related factors were regulated,and the inflammatory response was alleviated in rats with post-burn wound infections after the intervention with compound hibiscus flower and leaf extract.The mechanism might be related to the activation of Wnt/β-catenin signaling pathway.
