柴胡加龙骨牡蛎汤化裁对失眠小鼠的神经调节机制探索 被引量:4

摘要 目的研究柴胡加龙骨牡蛎汤化裁方对失眠小鼠脑干中5-羟色胺(5-hydroxytryptamine,5-HT)、5-羟基吲哚乙酸(5-hydroxyindole acetic acid,5-HIAA)、γ-氨基丁酸(γ-aminobutanoic acid,GABA)含量的影响。方法选取40只雄性小鼠随机分为模型组、柴胡组(柴胡加龙骨牡蛎汤治疗)、唑吡坦组(酒石酸唑吡坦片治疗)、空白对照组,每组10只。模型组、柴胡组、唑吡坦组(合称实验组)小鼠腹腔注射对氯苯丙氨酸造模,对照组腹腔注射生理盐水,共3天。造模成功后,对各组小鼠进行药物灌胃,柴胡组予柴胡加龙骨牡蛎汤化裁颗粒,唑吡坦组予酒石酸唑吡坦片,模型组和空白对照组予生理盐水。干预7天后取小鼠脑干,通过酶联免疫吸附法分别对5-HT、5-HIAA、GABA含量进行测定。结果模型组小鼠脑干中5-HT、5-HIAA、GABA的含量较空白对照组明显下降(P<0.05),说明造模成功;柴胡组与唑吡坦组小鼠脑干中5-HT、5-HIAA、GABA含量均高于模型组(P<0.05),但低于空白对照组(P<0.05);柴胡组与唑吡坦组小鼠脑干中5-HT、5-HIAA、GABA含量无明显统计学差异(P>0.05)。结论柴胡加龙骨牡蛎汤化裁方可改善失眠小鼠的睡眠质量,可能与其能够促进小鼠脑干中5-HT、5-HIAA、GABA的分泌有关,其短期疗效与酒石酸唑吡坦相当。 Objective To investigate the effects of bupleurum plus dragon bone and oyster shell decoction modified on the levels of 5-hydroxytryptamine(5-HT),5-hydroxyindole acetic acid(5-HIAA)andγ-Aminobutanoic acid(GABA)in the brain stem of insomnia mice.Methods 40 male mice were randomly divided into model group,bupleurum group(treated with bupleurum plus keel oyster decoction),zolpidem group(treated with zolpidem tartrate tablets)and blank control group,with 10 mice in each group.All mice in the model group,the bupleurum group and the zolpidem group(collectively referred to as the experimental group)were intraperitoneally injected with p-chlorophenylalanine(PCPA)to model for 3 days.In the meantime,the mice in the control group were intraperitoneally injected with physiological saline.After successful modeling,each mouse was given corresponding drugs by gavage for 7 days:the Bupleurum group was given dispensing granule of bupleurum plus dragon bone and oyster shell decotion modified,the zolpidem group was given zolpidem tartrate tablet,the model group and the control group were given physiological saline.A week of drug intervention later,all the brain stem in mice would be taken out to detect the levels of 5-HT,5-HIAA and GABA by the method of enzyme-linked immunosorbent assay(ELISA).Results The levels of 5-HT,5-HIAA and GABA in the model group were significantly lower than those in the control group(P<0.05).The levels of the bupleurum group and the zolpidem group were both higher than those in the model group(P<0.05),but lower than those in the control group(P<0.05).There was no statistically significant difference between the bupleurum group and the zolpidem group(P>0.05).Conclusion Bupleurum plus dragon bone and oyster shell decoction modified can improve the sleep quality of insomnia mice,which may be relevant to promote the secretion of 5-HT,5-HIAA and GABA in the brain stem of mice.Its short-term efficacy is almost equivalent to zolpidem tartrate.
