刺参早期发育阶段luqin型神经肽分布及表达特征研究

Localization and Expression Characteristics of luqin-Type Neuropeptide at Early Developmental Stages of Apostichopus japonicus

摘要 为探究神经肽在刺参(Apostichopus japonicus)幼体中的潜在功能,丰富对刺参神经生理学的认识。本研究利用免疫荧光和荧光定量技术初步探究了刺参的luqin型神经肽AjLQ及其前体基因AjLQP在刺参典型的早期发育阶段(囊胚期、原肠胚期、耳状幼虫期、樽形幼虫期和五触手幼虫期)的分布及表达特征。免疫荧光结果显示,在游泳、摄食和附着相关组织中观察到AjLQ阳性信号;荧光定量结果显示,在刺参幼体开始摄食的耳状幼虫期AjLQP基因的表达量显著高于其他早期发育阶段AjLQP基因的表达量。研究结果表明,luqin型神经肽是刺参幼体摄食、游泳和附着的潜在调控子。 In order to explore the potential function of neuropeptides in larvae of Apostichopus japonicus and enrich the knowledge about the neurophysiology of A.japonicus,in the present study,the localization and expression characteristics of luqin-type neuropeptide(AjLQ)and its precursor AjLQP in early developmental stages(blastula;gastrula,auricular,doliolaria and pentactula)of A.japonicus were detected using immunofluorescence and qRT-PCR.The results of immunofluorescence showed that positive signals of AjLQ were observed in swimming,feeding and adhesion related tissues.The results of qRT-PCR showed that the expression level of AjLQP at auricularia stage was significantly higher than that at other stages,which is the beginning of feeding for sea cucumber larvae.All these results suggested that luqin-type neuropeptide was a potential regulator of feeding,swimming and adhesion of A.japonicus larvae.
