摘要
目的探索中药治疗艾滋病免疫重建不良的用药规律及核心药物对免疫重建不良的作用机制。方法通过六大中英文数据库,包括中国医学生物文献数据库、万方数据库、中国期刊全文数据库、PubMed、Cochrane Library、Web of Science,检索从建库以来有关中药治疗艾滋病免疫重建不良的研究文献。利用IBM SPSS Statistics22.0及IBM SPSS Modeler 18.0软件进行数据分析挖掘。分析用药规律、核心药对,运用网络药理学手段研究其机制。结果纳入文献9篇,包含30味中药。频次前8味中药分别是黄芪、党参、灵芝、枸杞子、紫花地丁、人参、西洋参、冬虫夏草菌丝体,归经以肺、脾、肾为主,以甘、苦、辛味排前三,药性以平性、温性为主。核心药对组合是“灵芝-党参”,其主要通过7-甲氧基-2-甲基异黄酮、苍术内酯iii、果酮A、木犀草素、(1R)-2,3,4,9-四氢-1H-$b-咔啉-1-羧酸、黄豆黄素等成分作用于MAPK14、TNF、MAPK1、EGFR、AR、VEGFA等疾病靶点,调节免疫功能主要通过HIF-1、T cell receptor、Rap1、VEGF、MAPK等信号通路。结论中药治疗艾滋病免疫重建不良以补益肺脾肾为核心,通过多成分、多靶点、多途径对免疫重建不良发挥作用。
Objective To explore the medicinal mechanisms of traditional Chinese medicine(TCM)and core drugs in the treatment of AIDS immune reconstitution non-responders.Methods Six major Chinese and English databases,including the Chinese Medical Biological Literature Database,Wanfang Database,Chinese Journal Full Text Database,PubMed,Cochrane Library,and Web of Science,were used to retrieve literatures on TCM treatment for AIDS immune reconstitution non-responders since the establishment of the database.IBM SPSS Statistics 22.0 and IBM SPSS Modeler 18.0 software were used for data analysis and mining.Medicine rules and core drug pairs were analyzed,and network pharmacology methods were used to investigate the mechanism of action.Results Nine literatures were enrolled,including 30 TCM.The top 8 TCM in terms of frequency were Huangqi(astragali radix),Dangshen(codonopsis pilosula),Lingzhi(Ganoderma lucidum),Gouqizi(fruit of Chinese wolfberry),Zihuadiding(Chinese violet),Renshen(ginseng),Xiyangshen(American ginseng),Dongchongxiacao mycelium(cordyceps sinensis hyphas mycelium).The meridians are mainly focused on lung,spleen,and kidney,with the top three taste of sweet,bitter and pungent,respectively.The medicinal properties were mainly mild and warm.The core drug pair was Lingzhi(ganoderma lucidum)-Dangshen(codonopsis pilosula),which mainly exert on targets such as MAPK14,TNF,MAPK1,EGFR,AR,VEGFA and so on,through medicinal ingredients such as 7-Methoxy-2-methyl isoflavone,atractylenolide iii,frutinone A,luteolin,(1R)-2,3,4,9-tetrahydro-1H-$b-carboline-1-carboxylic acid,glycitein,etc.The immune function was mainly regulated through HIF-1,T cell receptor,Rap1,VEGF,MAPK and other signalling pathways.Conclusions The TCM treatment for AIDS immune reconstitution non-responders focuses on tonifying the lung,spleen and kidney,and plays a role through multi-ingredient,multi-target,and multi-path.
