一种潜在结核病特异性诊断分子标志物Circ-IRAKM的筛选鉴定

摘要 目的分析Circ-IRAKM在活动型结核病患者外周血中的表达及其与结核病患者临床特征的相关性,评价其作为活动型结核病分子诊断标志物的预测价值。方法通过公共数据库(Gene Expression Omnibus,GEO)筛选在结核病患者外周血差异表达的Circ-RNA,qRT-PCR技术验证Circ-IRAKM在活动型结核病患者外周血的相对表达,采用SPSS 23.0及ROC曲线等分析Circ-IRAKM作为诊断活动型结核病的价值,GO/KEGG数据库预测候选靶点生物学功能。结果Circ-IRAKM在活动型结核病患者外周血中升高有统计学意义(t=9.096,P<0.001);Circ-IRAKM在结核病患者外周血的AUC为0.966,特异性为0.995,灵敏度为0.937;且其表达与外周血巨噬细胞相对值/绝对值呈正相关;数据库预测其可能通过NF-κB信号通路参与病原-宿主博弈过程。结论Circ-IRAKM可作为结核病患者诊断及预后的候选特异性生物分子标志物。 Tuberculosis is a contagious disease caused by Mycobacterium tuberculosis and is one of the most serious threats to global public health.The screening of specific molecular markers of tuberculosis is important for rapid diagnosis and prognosis.In this study,Circ-RNA differentially expressed in peripheral blood of tuberculosis patients were sought by screening public databases(Gene Expression Omnibus,GEO).Several Circ-RNAs with differential expression in peripheral blood of tuberculosis patients were validated using qRT-PCR and agarose gel electrophoresis.It was found that Circ-IRAKM significantly increased in peripheral blood of active TB patients(t=9.096,P<0.001).The AUC of Circ-IRAKM in peripheral blood of tuberculosis patients was 0.966,the specificity was 0.995,and the sensitivity was 0.937.And the expression of Circ-IRAKM was positively correlated with the relative/absolute value of macrophages.GO/KEGG database predicted that Circ-IRAKM might participate in the pathogen-host game through NF-κB signaling pathway.Therefore,Circ-IRAKM has the potential as a specific biomarker for diagnosis and prognosis of TB patients.
