小柴胡汤对胆汁反流性胃炎模型大鼠相关胃肠激素及炎症因子的影响 被引量:3

Effects of Xiaochaihu Decoction(小柴胡汤)on related gastrointestinal hormones and inflammatory factors in model rats with bile reflux gastritis

摘要 目的 研究小柴胡汤对胆汁反流性胃炎模型大鼠相关胃肠激素及炎症因子的影响。方法 将60只SPF级SD大鼠采用随机数字表法分为空白组,模型组,阳性对照组,小柴胡汤低、中、高剂量组6组,每组10只。除空白组外,其余5组连续灌胃自制反流液建立胆汁反流性胃炎大鼠模型,空白组、模型组灌胃蒸馏水,其余4组灌胃相应药物进行干预。连续灌胃35 d,观察灌胃给药期间各组大鼠体质量变化,检测各组大鼠血清和胃黏膜瘦素(LEP)、胃泌素(GAS)、前列腺素E2(PGE2)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的含量,以及血浆和胃黏膜胃动素的含量。结果 模型组大鼠体质量增长缓慢,大鼠体质量明显低于空白组、阳性对照组及小柴胡汤高、中剂量组(P<0.05)。与模型组比较,阳性对照组、小柴胡汤高剂量组大鼠血清和胃黏膜LEP、GAS、PGE2含量明显升高,TNF-α、IL-1β含量明显降低,血浆和胃黏膜MTL含量明显升高,差异均有统计学意义(P<0.05或P<0.01);小柴胡汤中剂量组大鼠血清和胃黏膜LEP、PGE2含量明显升高,TNF-α、IL-1β含量明显降低,血清GAS含量和血浆MTL含量明显升高,差异均有统计学意义(P<0.05或P<0.01);小柴胡汤低剂量组大鼠血清和胃黏膜PGE2含量明显升高,TNF-α含量明显降低,胃黏膜LEP、GAS含量明显升高,血清IL-1β含量明显降低,差异均有统计学意义(P<0.05)。结论 小柴胡汤具有防治胃黏膜损伤及增强胃动力的作用,其干预胆汁反流性胃炎的作用机制与其可调节相关胃肠激素和炎症因子的含量有关。 Objective To study the effects of Xiaochaihu Decoction(小柴胡汤)on related gastrointestinal hormones and inflammatory factors in model rats with bile reflux gastritis.Methods 60 SPF SD rats were randomly divided into blank group,model group,positive control group,Xiaochaihu Decoction low,medium and high dose groups,with 10 rats in each group.Except the blank group,the other 5 groups were continuously gavaged with self-made reflux solution to establish the rat model of bile reflux gastritis.The blank group and model group were ga-vaged with distilled water,and the other 4 groups were gavaged with corresponding drugs for intervention.During 35 days gavage,the changes of body mass of rats in each group were observed,and the contents of leptin(LEP),gastrin(GAS),prostaglandin E2(PGE2),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in serum and gas-tric mucosa,as well as the contents of motilin(MTL)in plasma and gastric mucosa were detected.Results The body mass of rats in model group increased slowly,and the body mass of rats in model group was significantly lower than that in blank group,positive control group and Xiaochaihu Decoction high-dose and medium-dose groups(P<0.05).Compared with model group,the contents of LEP,GAS and PGE2 in serum and gastric mucosa were signifi-cantly increased,the contents of TNF-αand IL-1βwere significantly decreased,and the contents of MTL in plasma and gastric mucosa were significantly increased in positive control group and Xiaochaihu Decoction high-dose group,with statistical significance(P<0.05 or P<0.01).The contents of LEP and PGE2 in serum and gastric mucosa were significantly increased,the contents of TNF-αand IL-1βwere significantly decreased,and the contents of GAS in serum and MTL in plasma were significantly increased in Xiaochaihu Decoction medium-dose group,with statisti-cal significance(P<0.05 or P<0.01).The content of PGE2 in serum and gastric mucosa was significantly in-creased,the content of TNF-αwas significantly decreased,the contents of LEP and GAS in gastric mucosa were sig-nificantly increased,and the content of IL-1βin serum was significantly decreased in Xiaochaihu Decoction low-dose group,with statistical significance(P<0.05).Conclusion Xiaochaihu Decoction can prevent gastric mucosal injury and enhance gastric motility.The mechanism of its intervention in bile reflux gastritis is related to the regula-tion of the contents of related gastrointestinal hormones and inflammatory factors.
