HPLC同时测定明目益睛丸中5种有效成分含量

Simultaneous determination of five active components in Mingmu Yijing Pills by HPLC

目的:建立高效液相色谱法同时测定明目益睛丸中金丝桃苷、五味子醇甲、橙皮苷、毛蕊花糖苷、人参皂苷Rg1含量测定方法。方法:色谱柱Ultimate?ODS-3(4.6×250 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),流速为1.0 mL/min,检测波长为203 nm,柱温为30℃,进样量为10μL。结果:金丝桃苷、五味子醇甲、橙皮苷、毛蕊花糖苷、人参皂苷Rg1质量浓度分别在5.12~102.4μg/m L、5.00~100.0μg/mL、14.84~296.8μg/mL、5.56~111.2μg/mL、7.24~217.2μg/mL范围内与峰面积线性关系良好(r≥0.9995,n=6);精密度、稳定性、重复性试验的RSD均小于3%;平均加样回收率为96.96%~106.50%,RSD小于3%(n=6)。结论:该方法重复性良好、操作简便且稳定,可用于明目益睛丸有效成分的含量测定。

Objective:To establish a HPLC method for simultaneous determination of hyperoside,schizandrin,hesperidin,acteoside and ginsenoside Rg1 in Mingmu Yijing pills.Method:The determination was performed on Ultimate?ODS-3 (4.6×250mm,5μm) column with mobile phase consisted of acetonitrile-0.1%phosphoric acid aqueous solution (gradient elution) at the flow rate of 1.0 m L·min^(-1).The detection wavelength was set at 203 nm.The column temperature was 30℃and sample size was 10μL.Result:The mass concentrations of hypericin,schisandrin methyl,hesperidin,actinoside and ginsenoside Rg1 were 5.12~102.4μg·mL^(-1),5.00~100.0μg·mL^(-1),14.84~296.8μg·mL^(-1),5.56~111.2μg·mL^(-1),7.24~217.2μg·mL^(-1) respectively.There was a good linear relationship with the peak area (r≥0.9995,n=6).RSD of precision,stability and reproducibility tests were all less than 3%.The average recoveries were 96.96%~106.50%with RSD less than 3%(n=6).Conclusion:The method has good repeatability,simple operation and stability,and can be used for the determination of effective components in Mingmu Yijing pills.
