摘要
[目的]建立板蓝根中重金属元素的快速检测方法。[方法]采用电感耦合等离子体质谱法快速测定不同产地板蓝根中铅(Pb)、镉(Cd)、砷(As)、汞(Hg)、铜(Cu)的含量,了解河北、河南、安徽等不同产地板蓝根中5种重金属元素的差异,并结合主成分分析法和聚类分析法评价不同产地板蓝根药材质量差异性。[结果]5种重金属元素在各自范围内线性关系良好(R2≥0.999 6),平均加样回收率为97.63%~99.96%,RSD为1.43%~3.83%。30批样品中,86.67%样品Cd含量超标,26.67%样品Cu含量超标,66.67%样品未检出Hg,所采集样品中Hg、Pb、As均未超标。主成分分析提取2个成分,累计贡献率72.577%,Pb和Cd对板蓝根的质量评价有重要贡献。综合评分显示,采自河北安国板蓝根药材质量较好;聚类分析发现,不同产地的检测指标含量有明显差异。[结论]电感耦合等离子体质谱法可快速、准确测定板蓝根药材中5种重金属元素的含量,该方法高效准确,可用于板蓝根药材中重金属元素的质量控制。
[Objective]To establish a rapid detection method for heavy metal elements in Radix Isatidis.[Method]The content of Pb,Cd,As,Hg and Cu in Radix Isatidis from different regions was rapidly determined using inductively coupled plasma mass spectrometry(ICP-MS).The differences in five heavy metal elements in Radix Isatidis from different regions such as Hebei,Henan and Anhui were investigated,and the quality differences of Radix Isatidis from different regions were evaluated using principal component analysis and cluster analysis.[Result]There was a good linear relationship between the five heavy metals and harmful elements in their respective ranges(R 2≥0.9996).The average recoveries were 97.63%-99.96%,RSD was 1.43%-3.83%.In 30 batches of samples,86.67%of the samples had Cd content exceeding the standard,26.67%of the samples had Cu content exceeding the standard,66.67%of the samples had no Hg detected,and Hg,Pb,As in the collected samples did not exceed the standard.Two components were extracted by principal component analysis,with a cumulative contribution rate of 72.53%.Pb and Cd contributed significantly to the quality evaluation of Radix Isatidis.According to the comprehensive score,the quality of Radix Isatidis collected from Anguo,Hebei was good;cluster analysis showed that there were significant differences in the content of test indicators in different habitats.[Conclusion]ICP-MS can quickly and accurately determine the contents of five kinds of heavy metals in Radix Isatidis.This method is efficient and accurate,and can be used for the quality control of heavy metal elements in Radix Isatidis.
