提取方法对好氧颗粒污泥胞外聚合物组分的影响

摘要 采用超声-热提法和超声-阳离子交换树脂法(CER)提取好氧污泥颗粒化过程中不同层级的胞外聚合物(EPS),对比分析了反应器不同时期EPS的蛋白质和多糖质量分数、官能团和组分以及提取前后污泥的粒径分布。研究结果表明:超声可以有效提取不同性质污泥的疏松型EPS(LB-EPS)。热提法提取的TB-EPS质量分数相比CER法较多,但提取后余泥受热膨胀,粒径不降反增。无论采用何种提取方法,污泥颗粒化过程中TB-EPS的蛋白、多糖质量分数和二者比值的变化规律均一致。傅里叶红外光谱和三维荧光光谱分析显示,CER法提取的TB-EPS与二价阳离子关系更为密切,热提法所得的TB-EPS含有较多腐殖酸类物质。 Ultrasound-thermal extraction and ultrasound-cation exchange resin(CER)were used to extract different layers of extracellular polymers(EPS)from aerobic sludge granulation process.The protein and polysaccharide mass fractions,functional groups and fractions of EPS at different periods of the reactor and the granular size distribution of the sludge before and after extraction were compared and analyzed.The results showed that ultrasonication could effectively extract the lax EPS(LB-EPS)from sludge with different properties.The thermal extraction method extracted more TB-EPS compared to the CER method,but the residual sludge was thermally expanded after extraction and the particle size increased rather than decreased.The pattern of changes in the protein and polysaccharide mass fraction and the ratio of the two in TB-EPS during sludge granulation was consistent regardless of the extraction method used.Fourier infrared spectroscopy and three-dimensional fluorescence spectroscopy analysis showed that TB-EPS extracted by CER method was more closely related to divalent cations,and TB-EPS obtained by thermal extraction method contained more humic acid-like substances.
