基于紫外光电离-离子迁移谱分析呼出气体中三甲胺的方法研究

摘要 人体呼出气体中的三甲胺(TMA)可作为慢性肾病的潜力标志物。针对慢性肾病早期筛查的需求,基于紫外光电离-离子迁移谱(UV-IMS)发展了分析呼出气体中三甲胺的新方法。以丙酮作为掺杂剂,提升了三甲胺的电离效率;优化了电离源参数和漂移管温度;并考察了样品湿度对三甲胺定性及定量检测的影响。所建立方法对三甲胺的理论检出限为1.13×10^(-3) mg/m^(3),线性工作范围为1.32×10^(-2)~1.32×10^(-1) mg/m^(3)(相关系数为0.9916),平均相对标准偏差为1.84%。最后,通过呼出气体加标测试,验证了该方法在复杂基质下对三甲胺的检测能力。 Trimethylamine(TMA)in human exhaled breath is considered a potential marker for chronic kidney disease(CKD).A detection method of TMA in exhaled gas based on UV ionization-ion mobility spectrometry(UV-IMS)was developed for early screening of CKD.Firstly,the ionization efficiency of TMA was enhanced by using acetone as a dopant.Then,the ionization source parameters and drift tube temperature were optimized.Further,the effect of sample humidity on the qualitative and quantitative detection of TMA was also investigated.It is concluded that the theoretical detection limit of TMA is 1.13×10^(-3) mg/m^(3),the linear response range is 1.32×10^(-2)-1.32×10^(-1) mg/m^(3) and the mean relative standard deviation is 1.84%.Finally,the ability of the system to detect trimethylamine in complex matrices was validated by exhaled gas spiking tests.
