摘要
目的:在钛表面构筑有序微米凹坑结构并评价其成骨活性。方法:机械抛光纯钛片随机分为3组(n=3):A组对照组,未再做任何处理,B组用HF酸蚀处理,C组用NaCl-HF电化学刻蚀处理形成有序微米凹坑结构。将骨髓间充质干细胞(BMSCs)培养在样本表面。观察细胞形态,检测细胞黏附和细胞增殖,检测细胞胶原分泌和细胞外基质(ECM)矿化。结果:无序微米沟壑状结构的B组试样,其表面BMSCs倾向于多边形黏附形态,促进了细胞胶原分泌和ECM矿化(P<0.05)。20μm大小有序微米凹坑阵列的C组试样,其表面多边形黏附形态的BMSCs可跨越多个微米凹坑而更为铺展,并且伸出较多细长的伪足与周边的细胞呈现紧密连接,明显促进了细胞黏附、细胞胶原分泌和ECM矿化(P<0.05)。B组和C组表面BMSCs的细胞增殖虽然均受到了一定程度的抑制,但是其还是随着时间推移而明显增加。结论:有序微米凹坑结构的试样可以最大程度促进BMSCs的细胞黏附和成骨分化,并具有良好的成骨活性。
Objective:To fabricate ordered microholes on titanium surface and to study its effects on the osteogenic activity of bone marrow mesenchymal stem cells(BMSCs).Methods:Mechanically polished titanium samples were randomly divided into 3 groups(n=3).The samples of group A received no further treatment as the controls,those of group B were chemically etched by HF,and those of group C were electrochemically etched by NaCl-HF for the constrution of ordered microfossae on the samplde surface.BMSCs were culture on the sample surface.The cell morphology was observed,cell adhesion and proliferation were tested,the collagen secretion and extracellular matrix(ECM)mineralization were detected,respectively.Results:BMSCs on the samples of group B disordered microfossae exhibited polygonal morphologies,with slightly more collagen secretion and ECM mineralization(P<0.05).The polygonal BMSCs on the samples of Group C could span across multiple microfossae spreading more obviously and connecting closely with surrounding cells with noticeable pseudopodia extensions,more cell adhesion,more collagen secretion and more ECM mineralization(P<0.05).Although the proliferation of BMSCs in group B and C was slightly inhibited,but still significantly increased over time.Conclusion:The titanium samples with ordered microfossae may promote the BMSCs adhesion and osteogenic differentiation,and can support proliferation to some extent,and promote osteogenic activity of BMSCs.
