银叶金合欢枝多酚的提取工艺及抗氧化性研究 被引量:1

Extraction Technology and Antioxidant Activities of Polyphenols from Acacia Podalyriifolia Branches

摘要 以银叶金合欢枝为原料,探究提取溶剂、丙酮浓度、料液比、提取时间、提取温度对银叶金合欢枝多酚得率的影响,在此试验结果上设计4因素3水平的正交试验,确定银叶金合欢枝多酚提取的最佳工艺条件为:丙酮体积分数70%、料液比1:25(g/mL)、提取时间10 min、提取温度55℃,在此条件下,银叶金合欢枝多酚得率为15.21%。其次,探究银叶金合欢枝多酚提取液的体外抗氧化活性,测定提取液对·OH、DPPH·和·NO的清除效果,同时以维生素C作为阳性对照。当银叶金合欢枝多酚提取液质量浓度为2.50 mg/mL和1.0 mg/mL时,对·OH和·NO的清除率达到71.66%和57.33%,当银叶金合欢枝多酚提取液质量浓度为0.30 mg/mL时,对DPPH·的清除率达到90.96%,与维生素C清除效果相当,表明银叶金合欢枝多酚具有良好的抗氧化活性。 Using Acacia podalyriifolia branches as raw materials,the effects of extraction solvent,acetone concentration,solid-liquid ratio,extraction time,and extraction temperature on the extraction yield of polyphenols from Acacia podalyriifolia branches were explored.Based on the experimental results,four factors and three levels were designed.Orthogonal experiments were used to determine the optimal process conditions for the extraction of polyphenols from Acacia podalyriifolia branches as follows:70%acetone,solid-liquid ratio of 1:25(g/mL),extraction time of 10 min,and extraction temperature of 55℃,the yield of polyphenols from Acacia podalyriifolia was 15.21%.Secondly,the in vitro antioxidant activity of the crude polyphenol extract of Acacia podalyriifolia was investigated,and the scavenging effect of the extract on·OH,DPPH·and·NO was determined,and Vitamin C was used as a positive control.When the mass concentration of Acacia podalyriifolia polyphenol extract is 2.50 mg/mL and 1.0 mg/mL,the scavenging rates of·OH and·NO reached 71.66%and 57.33%,When the concentration of Acacia podalyriifolia polyphenol extract was 0.30 mg/mL,the scavenging rate of DPPH·reached 90.96%,which was comparable to the scavenging effect of Vitamin C,indicating that Acacia podalyriifolia polyphenols had good antioxidant activity.
