HPLC Determination of Adenosine in Acremonium Terricola Culture

运用HPLC法测地顶孢霉培养物中腺苷的含量,以期为地顶孢霉培养物的质量控制提供可靠的依据。结果本方法用于地顶孢霉培养物中腺苷的含量测定具有专属性;腺苷在0.4990~99.7976μg/mL范围内线性关系良好,相关系数R 2为1;腺苷峰面积精密度RSD为0.3%;腺苷的测定重复性RSD为0.2%;腺苷的回收率为97.09%~99.66%;耐用性较好。表明该方法线性范围宽、分析时间短、样品前处理简便、定量结果准确、重现性好,为其质量控制提供了依据。

HPLC method was used to determine the content of adenosine in Acremonium terricola culture in order to provide reliable basis for the quality control of Acremonium terricola culture.The results showed that the method was specific for the determination of adenosine in Acremonium terricola culture.The concentration of adenosine was in the range of 0.4990μg/mL to 99.7976μg/mL,and the correlation coefficient R 2 was 1.The precision RSD of adenosine peak area was 0.3%.The reproducibility RSD of adenosine was 0.2%.The recovery rate of adenosine was 97.09%~99.66%.The method had good durability.The results showed that the method had wide linear range,short analysis time,simple sample pretreatment,accurate quantitative results and good reproducibility,which provided a basis for its quality control.
