摘要
大部分山羊表现出季节性发情繁殖特点,但其分子遗传机理尚不清楚。近年来,越来越多的研究结果提示,哺乳动物的季节性繁殖与MTNR1A基因有关。为探明山羊MTNR1A基因的作用,采用荧光定量PCR(qRT-PCR)方法对常年发情的济宁青山羊和季节性发情的辽宁绒山羊下丘脑-垂体-卵巢繁殖轴组织中MTNR1A基因mRNA的表达差异进行了对比分析。组织表达谱结果发现,MTNR1A m RNA在两品种山羊的下丘脑、大脑皮层、海马体、卵巢和辽宁绒山羊垂体中均有表达。荧光定量PCR结果分析发现,MTNR1A基因在两品种山羊下丘脑中的表达量较高(P<0.05),在卵巢中有一定的表达,而在垂体中的表达量较低;在辽宁绒山羊下丘脑和卵巢中MTNR1A基因的表达量高于济宁青山羊(P>0.05)。提示MTNR1A基因可能与山羊季节性繁殖调控有关,值得进一步深入研究。
Most goat breeds represent seasonal reproduction,but its molecular genetic mechanism remains mysterious.Increasing data hinted a relationship between MTNR1A gene and seasonal reproduction in mammals recently.To unveil the role of MTNR1A gene in goat,the differences of mRNA expression in hypothalamus-pituitary-ovary axis(plus pineal and uterus)and genomic sequence structure between year-round estrous Jining Grey goat and seasonal Liaoning Cashmere goat were analyzed using quantitative real time polymerase chain reaction(qRT-PCR)and sequencing methods,respectively.The results showed that MTNR1A mRNA was mainly expressed in hypothalamus,cerebral cortex,hippocampus and ovary of both breeds,but just in Liaoning Cashmere goat for pituitary;the MTNR1A mRNA was expressed rather higher in hypothalamus(P<0.05),moderate in ovary and little lower in pituitary,and the expression was just little higher in Liaoning Cashmere goat than Jining Grey goat for hypothalamus and ovary(P>0.05).In the present study,the results suggested that MTNR1A gene might be involved in regulation of seasonal reproduction in goats,but further studies are needed.
