摘要
以花叶滇苦菜为原料,采用乙醇提取、静态吸附法筛选最适合花叶滇苦菜黄酮纯化的大孔树脂类型并优化纯化工艺。以颜色反应、薄层层析和紫外与红外扫描光谱鉴定黄酮类别。以DPPH自由基的清除率为评价指标,比较花叶滇苦菜黄酮纯化前后的抗氧化活性。结果表明,上样液p H3.0、浓度1.4 mg/mL、体积3 BV、40%乙醇为洗脱剂、洗脱体积4 BV、D101大孔树脂纯化后,黄酮纯度达56.02%,是纯化前的5.05倍。花叶滇苦菜黄酮类化合物中主要含有芦丁、木犀草素和槲皮素,极有可能含有芹菜素,共4类黄酮类化合物。纯化后花叶滇苦菜黄酮浓度小于0.3 mg/mL时,其对DPPH自由基清除率随其浓度的增大而增大,且高于二丁基羟基甲苯和维生素C;0.1 mg/m L浓度下其对DPPH自由基的清除率达86.64%,是纯化前的4.85倍。
Using Sonchus Asper as the raw material,ethanol extraction was carried out,and static adsorption method was used to screen the most suitable macroporous resin type for the purification of flavonoids from Sonchus Asper,and the purification process was optimized.The categories of flavonoids were identified by color reaction and thin layer chromatography,ultraviolet and infrared scanning spectroscopy.The scavenging rates of DPPH free radicals were used as evaluation indexes.The antioxidant activities of flavonoids from Sonchus asper were compared before and after purification.The results showed that D101 macroporous resin had good purification effect on the flavonoids ofSonchus asper.Under the conditions of pH 3.0,concentration 1.4 mg/mL,volume 3 BV,eluent 40%ethanol and elution volume 4 BV,the purity of flavonoids reached 56.02%,which was 5.05 times of that before purification.The flavonoids mainly contained rutin,luteolin and quercetin,and it was very likely to contain apigenin,there were 4 flavonoids in Sonchus asper.The scavenging rate of DPPH free radical was significantly enhanced.When the flavonoid concentration was less than 0.3 mg/mL,the scavenging rate of DPPH radical increased with the increase of flavonoid concentration,and was higher than that of dibutylhydroxytoluene and Vitamin C.The scavenging rate of DPPH free radical reached 86.64%at 0.1 mg/mL concentration,which was 4.85 times of that before purification.
