Siglecs家族分子在肿瘤中的表达与意义

摘要 唾液酸结合免疫球蛋白样凝集素(Siglecs)是一类唾液酸结合受体,在多种免疫细胞表面表达,具有复杂多样的免疫调节功能。Siglecs在肿瘤微环境(TME)中发挥不同的免疫及非免疫调节作用,介导炎症反应,促进肿瘤细胞发生免疫逃逸,调控肿瘤生长、侵袭、转移等生物学行为,加速肿瘤进展,影响患者预后与转归。因此,靶向Siglecs家族分子是目前肿瘤研究热点,并可能成为继PD-1之后肿瘤免疫治疗的明星分子。本文就Siglecs家族中影响肿瘤进展的分子类型、在TME中的作用机制及其作为治疗靶点的研究进展进行综述。 Sialic-acid-binding immunoglobulin-like lectins(Siglecs)are a family of sialic acid-binding receptors that are expressed on surface of a variety of immune cells and have complex and diverse immunomodulatory functions.Siglecs play different immune and non-immune regulatory roles in tumor microenvironment(TME),mediating inflammatory response,promoting immune escape of tumor and participate in tumor progression through various mechanisms such as regulating tumor growth and metastasis,and affecting prognosis and outcome of patients.Therefore,targeting Siglecs family molecules has aroused broad interest in current oncology research and may become a target therapeutic molecule of tumor immunotherapy,after PD-1.This paper reviews molecular types of Siglecs family that affect tumor progression,their mechanisms in TME and research progress as therapeutic targets.
