基于生物信息数据分析miR-655-3p在肝癌中的表达及其可能的调控机制

摘要 目的 探讨miR-655-3p在肝癌(liver hepatocellular carcinoma, LIHC)中的表达及其可能参与的生物学过程和信号通路,为进一步研究miR-655-3p的调控机制及生物学功能提供理论依据。方法 初步分析miR-655-3p在肝癌中的表达情况;借助多个数据库预测并筛选靶基因,随后对获取的靶基因进行GO功能富集和KEGG富集分析;构建靶蛋白互作网络并筛选网络中的核心基因,最后验证分析核心基因在肝癌中的表达及它们与临床分期、预后的关系。结果 miR-655-3p在肝癌组织中呈低表达,经筛选获得miR-655-3p相关靶基因153个。这些靶基因主要与RNA聚合酶II启动子转录调控、血管发育、衰老、凋亡调控等生物学过程以及神经营养因子、Rap1、MAPK等信号通路相关。蛋白互作网络中筛选的10个核心基因,仅RAC1表达水平在LIHC组织中显著上调,且与肝癌患者的临床分期和总生存期(overall surviral, OS)相关。结论 miR-655-3p在肝癌相关的生物学过程中起重要作用,可能与筛选的靶基因RAC1表达上调有关。 Objective To expore the expression and its possible biological processes and signaling pathways of miR-655-3p in hepatocellular carcinoma were discussed,and to provides theoretical basis for further study on the regulatory mechanism and biological function of miR-655-3p.Methods The expression of miR-655-3p in hepatocellular carcinoma tissues was preliminarily analyzed.Multiple databases were applied to predict and screen the target genes.Subsequently,GO functional enrichment and KEGG enrichment analysis were performed on the obtained target genes.Furthermore,the target protein interaction network was constructed and core genes in the network were screened.Finally,the expression of core genes in LIHC and their relationship with clinical stage and prognosis were verified and analyzed.Results miR-655-3p was low-expressed in hepatocellular carcinoma tissues.A total of 153 target genes related to miR-655-3p were screened in multiple databases.These target genes were mainly involved in transcriptional regulation of RNA polymerase II promoter,vascular development,aging,apoptosis regulation and other biological processes,as well as neurotrophin,Rap1,MAPK and other signaling pathways.Among the 10 core genes screened in the protein interaction network,only RAC 1 expression level was significantly upregulated in hepatocellular carcinoma tissues and correlated with clinical stage and overall survival(OS)of patients.Conclusion miR-655-3p plays an important regulatory role in the biological process related to hepatocellular carcinoma,which may be related to the upregulation of the screened target gene RAC 1 expression.
