北京信鸽中1株禽白血病病毒的分离和鉴定

摘要 为研究北京地区信鸽中禽白血病(AL)的流行情况,本试验从北京市沙河地区某市场抽样采集疑似AL信鸽血液样本共54份,通过检测p27抗原对其进行禽白血病病毒(ALV)鉴定;将分离株接种SPF鸡后检测其血清中ALV-Ab和ALV-J抗体,对分离株env基因进行扩增和测序,与各亚型ALV参考株gp 85基因核苷酸序列进行同源性分析,并构建系统发育进化树。结果显示,从54份样本中分离得到1株ALV,命名为CAU1501。ALV-J抗体检测全部呈阴性,ALV-Ab抗体检测有4份呈阳性。CAU1501的gp 85基因核苷酸序列与2007年美国分离株PDRC-3249同源性最高,与国内2009年山东分离株SDAU09E2遗传亲缘性最近,结合抗体检测结果确定分离株CAU1501为A亚群ALV。本试验结果可为后续北京地区鸽群AL的防控和净化提供参考依据。 In order to investigate the prevalence of avian leukosis(AL)in pigeons in Beijing area,this study collected a total of 54 suspected AL pigeon blood samples from a market in the Shahe district of Beijing.The samples were tested for p27 antigen to identify avian leukosis virus(ALV).The isolated strain was inoculated into SPF chickens,and their serum was tested for ALV-Ab and ALV-J antibodies.The env gene of the isolated strain was amplified and sequenced.Homology analysis was performed by comparing the nucleotide sequence of the gp 85 gene with various subtypes of ALV reference strains.The phylogenetic tree was constructed to analyze genetic relationships.The results showed that one strain of ALV,named CAU1501,was isolated from the 54 samples.All samples tested negative for ALV-J antibodies,while 4 samples tested positive for ALV-Ab antibodies.The nucleotide sequence of the gp 85 gene of CAU1501 exhibited the highest homology with the 2007 US isolate PDRC-3249 and the closest genetic affinity with the domestic 2009 Shandong isolate SDAU09E2,indicating that the isolated strain CAU1501 belongs to the A subgroup of ALV.The results of this study provide a reference for the prevention and purification of AL in pigeon populations in the Beijing area.
