针刺对脑梗死大鼠骨骼肌vimentin、desmin基因表达的影响

Effect of Acupuncture on the Expression of Vimentin and Desmin Gene in Skeletal Muscle of Cerebral Infarction Rats

摘要 目的观察针刺对脑梗死大鼠骨骼肌vimentin、desmin基因表达的影响,初步探讨针刺对脑梗死偏瘫大鼠骨骼肌的作用机制。方法将100只SD大鼠按随机数字表法分为:A组(头部穴位组)、B组(躯体穴位组)、C组(头部+躯体穴位组)、D组(模型组)、E组(空白组),每组20只,组内再按随机数字表法分为0、24、48 h组,每组5只。采用大脑中动脉线栓法制备大鼠脑缺血模型,将成功建立的大鼠模型,随机二次分组,每组20只,正常喂养。A组:针刺大鼠的头部穴位:百会、风府;B组:针刺大鼠躯体穴位后会、环跳、前三里、阳陵泉、后三里。C组:针刺大鼠的百会、风府、后会、环跳、前三里、阳陵泉、后三里。以上三组均针刺一次,捻转1min,平补平泻,留针30min;D组:造模后不施加任何刺激;E组:空白对照不造模,正常喂养,以上2组不予针刺。按时相在规定的骨骼肌部位(前肢肌、腓肠肌)取材并标记,采用qRT-PCR检测,得出不同时相针刺后的vimentin、desminm的mRNA表达变化情况。结果在实验中,所取骨骼肌部位vimentin及desmin的mRNA表达均出现显著差异(P<0.05),其中,在前肢肌vmentin的mRNA表达中,C组在0、24 h时表达量呈较明显上调趋势。在前肢肌及腓肠肌vimentin、前肢肌及腓肠肌desmin的mRNA表达中,B组表达量在0、24 h时均呈现明显上调。A组在前肢肌vimentin、前肢肌desmin、腓肠肌desmin的mRNA表达量中,在0、24、48 h均呈现上调。D组较A组、B组、C组的前肢肌vimentin、腓肠肌vimentin、前肢肌desmin、腓肠肌desmin的mRNA表达量在48 h时呈现明显上调。结论实验结果提示针刺改善脑梗死偏瘫骨骼肌肌力的机制可能是通过针刺对外周效应器的局部刺激,在改善局部功能的同时,引起中枢脑组织的修复,从而增强了外周效应器的康复,证实外周-中枢-外周的部分作用机制。 Objective:To observe the effect of acupuncture on the expression of vimentin and desmin genes in skeletal muscle of cerebral infarction rats and to explore the mechanism of acupuncture on skeletal muscle of cerebral infarction rats with hemiplegia.Methods:100 SD rats were divided into group A(head acupoint group),group B(body acupoint group),group C(head+body acupoint group),group D(model group)and group E(blank group)according to random number table method,20 rats each group,and then divided into 0,24 and 48 hours groups according to random number table method,5 rats each group.The cerebral ischemia rat model was prepared by middle cerebral artery suture method.The rat models were randomly sub-divided into two groups with 20 rats each group and fed normally.Group A was given acupuncture at head points,Baihui and Fengfu.Group B given acupuncture at body points,Houhui,Huantiao,Qiansanli,Yanglingquan and Housanli.Group C given acupuncture at head and body points,Baihui,Fengfu,Houhui,Huantiao,Qiansanli,Yanglingquan and Housanli.The above three groups were all needled once,twirling for 1 min,mild reinforcing and attentuating,retaining the needles for 30 min.Group D had no stimulus applied after molding.Group E had no modeling in the blank control and was normally fed.The above groups had no acupuncture.Samples were taken from the specified skeletal muscle(forelimb muscle and gastrocnemius muscle)and labeled on time,and qRT-PCR was used to detect the mRNA expression changes of vimentin and desminm after acupuncture at different times and phases.Results:In this experiment,the mRNA expressions of vimentin and desmin in the skeletal muscle showed significant differences(P<0.05),among which,in the mRNA expression of vmentin in forelimb muscle,the expression levels of group C showed a significantly up-regulated trend at 0 and 24 hours.And in the mRNA expression of vimentin and desmin in the forelimb muscle and gastrocnemius muscle,the expression levels of group B were significantly up-regulated at 0 and 24 hours.The mRNA expression levels of vimentin and desmin in gastrocnemius muscle in group A were up-regulated at 0,24 and 48 hours.Compared with that of groups A,B and C,the mRNA expression levels of forelimb muscle vimentin,gastrocnemius vimentin,forelimb muscle desmin and gastrocnemius desmin of group D were significantly up-regulated at 48 hours.Conclusion:The experimental results suggest that the mechanism of acupuncture improving the muscle strength of hemiplegic skeletal muscle in cerebral infarction may be through local stimulation of the peripheral effector by acupuncture,which not only improves the local function,but also causes the repair of the central brain tissue,thus enhancing the rehabilitation of the peripheral effector and confirming the partial mechanism of action of the peripheral,central and peripheral.
