长链非编码RNA FBXL19-AS1在胃癌中的作用机制研究

摘要 目的:探讨长链非编码RNA(Long non-coding RNA,lncRNA)FBXL19-AS1在胃癌中的作用机制。方法:癌症基因组图谱数据库提取胃癌表达谱数据;R语言筛选差异RNA,构建竞争内源性RNA(competing endogenous RNA,ceRNA)网络;R语言评估对胃癌患者生存的影响;ROC曲线预测对胃癌的诊断价值。结果:获得1004个差异lncRNA、104个微小RNA(MicroRNA,miRNA)、1641个信使RNA(Messenger RNA,mRNA);构建包含64个lncRNA、10个miRNA和10个mRNA的ceRNA网络,其中10个lncRNA,2个mRNA和1个miRNA对胃癌的预后有显著意义;筛选出胃癌相关的竞争内源性RNA轴,即F-box和富含亮氨酸的重复蛋白19反义RNA1(F-box and leucine-rich repeat protein 19 antisense RNA1,FBXL19-AS1)/miR-431/E盒锌指结合蛋白1(zinc finger E-box binding homeobox 1,ZEB1),三者的ROC曲线下面积均为0.8以上。结论:FBXL19-AS1可通过对miR-431/ZEB1轴的调节在胃癌中发挥致癌作用,靶向FBXL19-AS1可能为胃癌的治疗提供新策略。 OBJECTIVE To investigate the mechanism of action of Long non-coding RNA(lncRNA)FBXL19-AS1 in gastric can⁃cer(GC).METHODS The Cancer Genome Atlas(TCGA)database was used to extract the expression profile data of gastric cancer;Screening differential RNA using R and constructing competitive endogenous RNA(ceRNA)networks;Evaluate the impact on the sur⁃vival of GC patients using R.Predicting the diagnostic value of GC by ROC.RESULTS 1004 differential lncRNAs,104 microRNAs(miRNAs),and 1641 messenger RNAs(mRNA)were obtained.Construct a ceRNA network consisting of 64 lncRNAs,10 miRNAs,and 10 mRNAs.Among them,10 lncRNAs,2 mRNAs,and 1 miRNA have significant prognostic implications for GC.Screening out the competitive endogenous RNA axis related to gastric cancer,namely FBXL19-AS1/miR-431/ZEB1,all with ROC curve areas above 0.8.CONCLUSION FBXL19-AS1 can play a carcinogenic role in gastric cancer by regulating the miR-431/ZEB1 axis,and targeting FBXL19-AS1 may provide a new strategy for the treatment of gastric cancer.
