基于文献计量的多黏菌素E治疗药物监测相关研究热点和未来趋势分析

摘要 目的基于Web of Science数据库,利用文献计量学和可视化分析探索多黏菌素E治疗药物监测(TDM)相关研究的现状、热点和发展趋势。方法收集Web of Science核心数据库2012至2022年收录的多黏菌素E TDM相关研究的文献,应用机器学习和可视化工具,包括VOSviewer软件、书目共现分析系统和图形聚类工具包进行文献计量学分析。结果本研究共纳入713篇文献,2012至2022年文献的发表量有明显上升。在该领域贡献最多的国家和研究者分别是美国和LI J团队。多黏菌素E是出现最多的关键词,并与肾毒性和药物代谢动力学关联度最高。研究领域包括了多黏菌素E剂量与肾毒性的关系、联合给药的抗菌活性及耐药性、治疗监测与药代动力学。群体药代动力学和相关的模型是最新的研究热点。结论本研究文献计量分析能更好地了解多黏菌素E TDM相关研究的现状,并对未来研究进行了展望,未来应加快推进多黏菌素E TDM室间质控和评价研究,完善质控方法。此外应对硫酸黏菌素群体药代动力学及剂量优化进行更多的探索研究。 Objective Based on the Web of Science database,the current situation,hotspots and development trends of polymyxin E therapeutic drug monitoring(TDM)were explored by bibliometrics and visual analysis.Methods The literatures related to polymyxin E TDM included in the core database of Web of Science from 2012 to 2022 were collected,machine learning and visualization tools,including VOSviewer software,bibliographic co-occurrence analysis system and graphical clustering toolkit were applied for bibliometric analysis.Results A total of 713 publications were analyzed,from 2012 to 2022,research output increased significantly.In this field,the countries that have made the most contributions are the United States,and the most prolific researcher was LI J group.Polymyxin E was the most frequent keyword,with strong links to nephrotoxicity and pharmacokinetics.Study areas included relationship between polymyxin E dosage and nephrotoxicity,antimicrobial activity and resistance of combination therapy,as well as treatment monitoring and pharmacokinetics.Population pharmacokinetics and associated models constitute the latest research hotspots.Conclusions This bibliometric analysis will better understand the research status of TDM research related to polymyxin E and forecast future research trends.Priority should be given in promoting inter-laboratory quality control and evaluation studies for polymyxin E TDM,thus refining the quality control methods.Additionally,more extensive exploratory research is warranted concerning population pharmacokinetics and dose optimization of colistin sulfate.
