青年男性肥胖相关基因单核苷酸多态性与精液质量参数的相关性分析

Correlation analysis between single nucleotide polymorphisms of the fat mass and obesity associated gene and semen quality parameters in young males

摘要 目的通过对青年男性肥胖相关基因(FTO基因)单核苷酸rs9930609多态性进行检测,探讨其对精液质量的影响,间接判断其是否是影响男性生育力的危险因素。方法选取2021年1月至12月在甘肃省妇幼保健院进行备孕前检查的122例肥胖青年男性与120例非肥胖青年男性作为研究对象。检测其精液质量及外周血FTO基因rs9930609(A/T)多态性,分析FTO基因单核苷酸多态性与精液质量参数变化的关系。结果AA型基因携带者的体重、体重指数(BMI)、精子密度、精子DNA碎片率(DFI)与AT及TT型基因携带者比较,差异具有统计学意义(P<0.05);精子密度<15×10~6/mL人群的AA型基因携带率为22.03%,高于精子密度≥15×10~6/mL人群13.66%的AA型基因携带率(P=0.022);DFI>15%人群的AA型基因携带率为30.28%,显著高于DFI≤15%人群11.94%的AA型基因携带率(P=0.002);AA型基因携带者发生肥胖的风险是TT型基因携带者的5.834倍(P=0.000);A等位基因携带者发生肥胖的风险是T等位基因携带者的3.832倍(P=0.003)。结论FTO基因rs9939609(A/T)多态性改变与精液参数具有相关性,AA型等位基因所介导的肥胖是导致精子密度异常、DFI异常的重要诱因,也是间接影响男性生育力的危险因素。对于存在精液质量异常的肥胖男性,检测其FTO基因rs9939609(A/T)多态性,将有助于制订更为有效的减肥措施,从而改善精子质量。 Objective By detecting the single nucleotide rs9930609 polymorphism of fat mass and obesity associated gene(FTO gene)in young men,this paper aim to explore its impact on semen quality and indirectly determine whether it is a risk factor affecting male fertility.Methods A total of 122 obese young men and 120 non⁃obese young men who underwent pre⁃pregnancy examinations at Gansu Provincial Maternity and Child⁃care Hospital from January to December 2021 were selected as the research subjects.Their semen quality and peripheral blood FTO gene rs9930609(A/T)polymorphism were detected,and the correlation between FTO gene single nucleotide polymorphism and semen quality parameters was analyzed.Results The body weight,body mass index(BMI),sperm density,and sperm DNA fragmentation index(DFI)of AA type gene carriers were compared with those of AT and TT type gene carriers,and the differences were statistically significant(P<0.05).The carrying rate of AA type genes in individuals with sperm density<15×10^(6)/mL was 22.03%,which was higher than the carrying rate of AA type genes in individuals with sperm density≥15×10^(6)/mL of 13.66%(P=0.022).The carrying rate of AA type genes in individuals with DFI>15%is 30.28%,which is significantly higher than the carrying rate of AA type genes in individuals with DFI≤15%,which is 11.94%(P=0.002).The risk of obesity among AA genotype carriers is 5.834 times higher than that of TT genotype carriers(P<0.001).The risk of obesity among A allele carriers is 3.832 times higher than that among T allele carriers(P=0.003).Conclusions The polymorphism of the FTO gene rs9939609(A/T)is correlated with semen parameters.Obesity mediated by the AA type allele is an important cause of abnormal sperm density and DFI,and also an indirect risk factor affecting male fertility.For obese men with abnormal semen quality,testing their FTO gene rs9939609(A/T)polymorphism will help develop more effective weight loss measures and improve sperm quality.
