阿纳其根醇提物对衰老小鼠认知障碍改善作用

摘要 目的:研究阿纳其根醇提物对D-半乳糖联合三氯化铝(AlCl_(3))致痴呆小鼠模型学习记忆及抗氧化作用的影响。方法:采用腹腔注射D-半乳糖联合灌胃AlCl_(3)建立痴呆模型。采用跳台试验测试小鼠学习记忆能力;采用酶联免疫吸附法(ELISA)检测小鼠血清、海马组织和大脑皮层中脂褐素和活性氧(ROS)的含量;采用比色法检测超氧化物歧化酶(SOD)的活性。结果:跳台试验中,与模型组比,阿纳其根醇提物各剂量组潜伏期延长,错误次数有降低趋势;在生化指标检测中,阿纳其根醇提物各剂量组可减少小鼠海马组织中脂褐素、ROS含量,提高海马组织和大脑皮层中SOD活性。结论:阿纳其根醇提物能够改善D-半乳糖联合AlCl_(3)致痴呆小鼠模型学习记忆障碍,其作用机制可能与其提高体内抗氧化能力,减少脑组织之中脂褐素的堆积有关。 Objective:To investigate the effect of Anacyclus pyrethrum alcohol extract on the learning record and antioxidant effect of D‑galactose combined with AlCl_(3)in dementia mouse model.Methods:A dementia model was established by intraperitoneal injection of D‑galactose combined with gavage of AlCl_(3).Step‑down test was tested the learning and memory ability of mice;The contents of lipofuscin and reactive oxygen species(ROS)in serum,hippocampus and cortex of mice were detected by enzyme‑linked immunosorbent assay(ELISA);And the activity of superoxide dismutase(SOD)was detected by colorimetry.Results:In the step‑down test,compared with the model group,the incubation period of each Anacyclus pyrethrum alcohol extract group was significantly prolonged,and the number of mistakes tends to decrease.In the detection of oxidative stress indicators,the alcohol extract of Anacyclus pyrethrum in each dose group could significantly reduced lipofuscin and ROS in hippocampus,and significantly increase SOD in hippocampus and cortex.Conclusion:The Anacyclus pyrethrum alcohol extract could improve the learning and memory impairment of D‑galactose combined with AlCl_(3)in mice with chronic cognitive impairment,and the mechanism of action might be related to improving the antioxidant capacity in the body and reducing the accumulation of lipofuscin in the brain tissue.
