Notch信号通路调控T细胞免疫在气道炎症性疾病中的作用

摘要 气道炎症性疾病发病机制复杂,T淋巴细胞是气道炎症的重要免疫屏障。研究表明Notch信号通路与T细胞免疫功能关系密切,两者间的调节作用对于气道炎症性疾病的发生和发展具有显著影响,近年来相关研究已取得了显著进展。Notch信号通路参与多种T细胞的发育、分化和活化过程,维持细胞生存并增强杀伤能力;通过调控T细胞平衡状态,调节细胞因子表达,促进气道炎性细胞浸润、上皮细胞化生和气道黏液分泌等途径,参与气道炎症性疾病的病理学改变。针对Notch通路上的靶点,相关治疗药物研究也取得了一定进展,包括γ-分泌酶抑制剂、树突状细胞免疫疗法以及中药等在不同气道炎症疾病模型中已被证明有效。本文就Notch信号通路调控T细胞免疫与气道炎症性疾病的相关研究进行综述,以期为临床治疗气道炎症性疾病提供新的突破口。 The pathogenesis of inflammatory airway diseases is complex.T lymphocytes construct important immune barriers for airway inflammation.Results of related studies demonstrated that Notch signaling pathway is closely related to immune function of T cells,while regulation between them has a significant impact on occurrence and development of inflammatory airway diseases.Significant progress has been achieved in this field in recent years.Notch signaling pathway is involved in development,differentiation and activation of various subtypes of T cells,maintaining cell survival and enhancing kill capability of cells.Notch signal play important roles in pathological changes of inflammatory airway diseases,through regulation of T cell balance status,expression of cytokines,promotion of airway inflammatory cell infiltration,epithelial cell metaplasia and mucus secretion of airway.Effectiveness of some drugs,includingγ-secretory enzyme inhibitors,dendritic cell immunotherapy and traditional Chinese medicine had been demonstrated for different targets of Notch signaling pathway among different inflammatory airway diseases.Researches on the field of Notch signaling pathway regulated T cell immunity in inflammatory airway diseases were reviewed in this article,providing novel breach for treatment in clinical practice.
