基于核桃油-辛酸酶法合成新型结构脂的研究

摘要 研究以核桃油、辛酸为实验原料,通过Box-Benhnken响应面优化核桃油-辛酸结构脂的制备工艺,以辛酸插入率为评价标准,从固定化脂肪酶Novozym 435、Lipozyme TL IM和Lipozyme RM IM中挑选出辛酸插入率最高的酶,考察底物摩尔比、酶的添加量、反应时间及反应温度对核桃油-辛酸结构脂中辛酸插入率的影响,在单因素实验的基础上,利用四因素三水平响应面法确定核桃油-辛酸结构脂的制备工艺。结果表明固定化脂肪酶Lipozyme TL IM较适合制备核桃油-辛酸结构脂,其最佳酸解反应的条件为底物摩尔比(辛酸∶核桃油)为3∶1,酶质量分数为10%(质量分数),反应时间为12.6 h,反应温度为44.8℃,在此条件下,辛酸插入率为32.37%。 In this paper,walnut oil and octanoic acid were used as experimental raw materials,and the preparation process of walnut oil octanoic acid structured lipids was optimized by Box-Benhnken response surface.The enzyme with the highest octanoic acid insertion rate was selected from the immobilized lipases Novozym 435,Lipozyme TL IM and Lipozyme RM IM based on its octanoic acid insertion rate.The molar ratio of substrate,the amount of enzyme added the effects of reaction time and temperature on the insertion rate of octanoic acid in walnut oil octanoic acid structured grease were studied.On the basis of single factor test,the preparation process of walnut oil octanoic acid structured grease was determined by four-factor three-level response surface method.The results indicated that the immobilized lipase Lipozyme TL IM was more suitable for the preparation of walnut oil octanoic acid structured lipids.The optimum acidolysis conditions were as follows:the molar ratio of substrate(octanoic acid∶walnut oil) was 3∶1,the mass fractiom enzyme added was 10%,the reaction time was 12.6 h,the reaction temperature was 44.8 ℃,and the insertion rate of octanoic acid was 32.37%.These condition were suitable for the preparation of walnut oil octanoic acid structured grease.
