摘要
利用串联质谱标签(Tandem Mass Tag,TMT)标记定量蛋白质组学技术对水牛初乳和常乳乳清蛋白进行蛋白质组分析,筛选得到具有相关免疫功能的差异蛋白质49种。对这些免疫性差异蛋白进行生物信息学分析发现,其主要参与的生物过程为免疫系统过程、防御反应、应激反应;主要参与的细胞组分为胞外区域、血液微粒;主要参与的分子功能为结合受体;主要参与的代谢通路为补体和凝血级联、产生IgA的肠道免疫、造血细胞系和NF kB信号通路等免疫和炎症相关通路。通过蛋白质互作网络分析,找到一些具有高连接度的免疫蛋白因子。本研究为开发增强免疫活性的高端水牛乳制品提供了一定参考。
The quantitative proteomics technique of Tandem Mass Tag(TMT)was used to analyze the proteome of buffalo colostrum and normal milk whey protein proteins,and 49 differential proteins with related immune functions were screened.Bioinformatics analysis of these immune differentially expressed proteins revealed that they mainly participate in biological processes such as immune system processes,defense responses,and stress responses;The main cell components involved were extracellular region and blood particles;the main molecular function involved was binding receptor;the main metabolic pathways were complement and coagulation cascade,IgA-producing intestinal immunity,hematopoietic cell line and NF-kB Signaling pathways related to immunity and inflammation.Identify some immune protein factors with high connectivity through protein interaction network analysis.This study provides a certain reference for the development of high-end buffalo dairy products that enhance immune activity.
