肝素化脱细胞血管支架的制备及血液相容性评价

摘要 背景:脱细胞血管支架能模仿天然血管的微观结构与功能,但在制备过程中出现部分细胞外基质丢失,导致其血液相容性受到影响,因此有必要对脱细胞血管支架进行修饰以提升其血液相容性。目的:评价经Triton-x100/肝素钠处理制备的脱细胞血管支架的血液相容性。方法:从SD大鼠中取腹主动脉,随机分为对照组与实验组,对照组采用Triton-x100进行脱细胞处理48 h,实验组采用Triton-x100进行脱细胞处理48 h后进行肝素钠修饰处理。检测两组脱细胞血管支架的形貌、亲水性能,通过复钙化凝血时间实验、血小板黏附实验、动态凝血时间实验、溶血实验及补体激活实验评估两组脱细胞血管支架的血液相容性。结果与结论:①扫描电镜显示两组血管支架表面相对完整,脱细胞处理后血管支架表面出现大量纤维丝,表面微观结构发生了较明显改变;两组血管支架的水接触角均小于天然血管(P<0.0001),两组血管支架的水接触角比较差异无显著性意义(P>0.05)。②实验组血管支架的复钙化凝血时间长于对照组(P<0.05),血管支架膜上黏附的血小板数量少于对照组(P<0.0001),凝血指数大于对照组(P<0.01),补体水平低于对照组(P<0.001);两组血管支架的溶血率均低于国家标准5%。③结果表明,经Triton-x100/肝素钠处理的血管支架具有优良的血液相容性。 BACKGROUND:Acellular vascular scaffolds can mimic the microstructure and function of native blood vessels,but some extracellular matrix loss occurs during their preparation,which affects their hemocompatibility.Therefore,it is necessary to modify them to improve their hemocompatibility.OBJECTIVE:To assess the hemocompatibility of acellular vascular scaffold prepared by Triton-x100/heparin sodium treatment.METHODS:The abdominal aorta was taken from SD rats and randomly divided into control and experimental groups.The control group was treated with Triton-x100 for 48 hours.The experimental group was treated with Triton-x100 for 48 hours and then treated with heparin sodium.The morphology and hydrophilicity of the two groups of acellular vascular scaffolds were detected.The hemocompatibility of the two groups of acellular vascular scaffold was evaluated by recalcification coagulation time test,platelet adhesion test,dynamic coagulation time test,hemolysis test,and complement activation test.RESULTS AND CONCLUSION:(1)Scanning electron microscopy showed that the surface of the two groups of vascular scaffolds was relatively intact,and a large number of fiber filaments appeared on the surface of the scaffolds after decellularity treatment,and the surface microstructure changed significantly.The water contact angle of the two groups of vascular scaffolds was smaller than that of natural vessels(P<0.0001).There was no significant difference in water contact angle between the two groups(P>0.05).(2)The coagulation time of vascular scaffold was longer in the experimental group than in the control group(P<0.05).The number of platelets attached to the scaffold membrane was less in the experimental group than that in the control group(P<0.0001).The coagulation index was greater in the experimental group than that in the control group(P<0.01),and the complement level was lower in the experimental group than that in the control group(P<0.001).The hemolysis rate of the two groups was lower than 5%of the national standard.(3)To conclude,acellular scaffold treated with Triton-x100/heparin sodium has excellent hemocompatibility.
