Expression and significance of jumonji domain-containing protein 2B and hypoxia inducible factor-1αin non-Hodgkin lymphoma tissues in children

目的探讨含jumonji结构域的蛋白2B(jumonji domain-containing protein 2B,JMJD2B)和缺氧诱导因子1α(hypoxia inducible factor-1α,HIF-lα)在儿童非霍奇金淋巴瘤(non-Hodgkin's lymphoma,NHL)组织中的表达和意义。方法应用免疫组化检测46例NHL儿童(观察组)和24例反应性增生儿童(对照组)淋巴结组织标本中JMJD2B和HIF-1α的表达,分析JMJD2B和HIF-1α与NHL患儿临床病理特征和预后的关系,以及在NHL组织中JMJD2B和HIF-1α表达的相关性。结果观察组JMJD2B(87%vs 21%)和HIF-1α(83%vs 42%)表达阳性率均高于对照组(P<0.05)。JMJD2B和HIF-1α表达与NHL患儿血清乳酸脱氢酶水平和国际预后指数危险度相关(P<0.05)。JMJD2B与HIF-1α在NHL患儿组织中的表达呈正相关(rs=0.333,P=0.024)。结论JMJD2B和HIF-1α在儿童NHL组织中表达增高,二者在儿童NHL发生发展过程中可能起协同作用;JMJD2B可作为儿童NHL辅助诊断、评估病情、指导治疗、评估预后的新指标.

Objective To investigate the expression and significance of jumonji domain-containing protein 2B(JMJD2B)and hypoxia-inducible factor-1α(HIF-1α)in non-Hodgkin's lymphoma(NHL)tissues in children.Methods Immunohistochemistry was used to detect the expression of JMJD2B and HIF-1αin lymph node tissue specimens from 46 children with NHL(observation group)and 24 children with reactive hyperplasia(control group).The relationship between JMJD2B and HIF-1αexpression with clinicopathological characteristics and prognosis in children with NHL,as well as the correlation between JMJD2B and HIF-1αexpression in NHL tissues,were analyzed.Results The positive expression rates of JMJD2B(87%vs 21%)and HIF-1α(83%vs 42%)in the observation group were higher than those in the control group(P<0.05).The expression of JMJD2B and HIF-1αwas correlated with serum lactate dehydrogenase levels and the risk of international prognostic index in children with NHL(P<0.05).The expression of JMJD2B was positively correlated with the HIF-1αexpression in children with NHL(rs=0.333,P=0.024).Conclusions JMJD2B and HIF-1αare upregulated in children with NHL,and they may play a synergistic role in the development of pediatric NHL.JMJD2B can serve as a novel indicator for auxiliary diagnosis,evaluation of the severity,treatment guidance,and prognosis assessment in pediatric NHL.
