纳米黑炭经气管染毒对小鼠肝脏的影响 被引量:1

摘要 我国每年因大气颗粒物造成大量人口死亡和经济损失,研究其对肝脏造成毒性影响,为大气颗粒物对人群健康的危害研究提供借鉴意义。纳米黑炭模拟大气颗粒物对小鼠进行染毒,按照纳米黑炭与小鼠体重4 mg/kg的比例,每只小鼠气管滴注量为50μL,分组为处理1 d、3 d、7 d(每天染毒1次)和空白对照组4组,通过HE染色制做肝脏组织病理学切片,对切片进行病理观察。并对其余肝脏,测定碱性磷酸酶(AKP)、酸性磷酸酶(ACP)的活性。染毒1 d,可见大量纳米黑炭颗粒在血管中,大量炎细胞浸润;染毒3 d,炎细胞浸润,肝细胞结构异常,排列紊乱;染毒7 d,肝细胞气球样变。随着纳米黑炭暴露的时间延长会引起碱性磷酸酶和血清酸性磷酸酶的活性发生变化,但最终显示降低。纳米黑炭破坏肝脏的代谢平衡,并且纳米黑炭暴露的时间越长,肝脏产生的损伤和病变越严重。 Atmospheric particulate matter causes a large number of deaths and economic losses every year in our country.So the study on its toxic effects on the liver can provide reference for the harm of atmospheric particulate matter to human health.According to the ratio of nano-black carbon to mice's body weight of 4 mg/kg,nano-black carbon simulated atmospheric particles were poisoned to mice.Each mouse was given 50μL by intratracheal instillation.The mice were divided into four groups:treatment for 1,3 and 7 days(poi⁃soned once a day)and blank control group.Liver histopathological sections were made by HE staining,and pathological observations were made on these sections.Additionally,the activity changes of alkaline phospha⁃tase(AKP)and acid phosphatase(ACP)in livers were measured.After 1 day of poisoning,a large number of black carbon nanoparticles could be observed in the blood vessels,and a large number of inflammatory cells were infiltrated.After 3 days of poisoning,inflammatory cells were infiltrated,and the structure of liver cells was abnormal and disordered.After 7 days of poisoning,ballooning degeneration of hepatocytes were ob⁃served.The exposure time of nano black carbon caused changes in the activity of AKP and ACP,but eventual⁃ly followed by a decrease.Nano black carbon destroyed the metabolic balance of liver,and the longer the ex⁃posure time of nano black carbon,the more serious the liver damage and lesions.
