摘要
咔唑类生物碱(morindolestatin)源自链霉菌HUZ0001,其结构独特,具有优异的杀菌、抗氧化活性.为提高morindolestatin的产量,以链霉菌HUZ0001为试验菌株,采用单因素试验与正交设计实验相结合的方法,对培养基种类、发酵时间、接种量、装液量、发酵温度和转速等发酵条件进行优化.结果表明,优化的发酵培养基为:2.0%可溶性淀粉、1.0%棉籽饼粉、0.5%酵母抽提物、0.5%麦芽提取物、0.2%MgSO_(4)·7H_(2)O、0.2%NaCl、0.2%CaCO_(3).优化的发酵条件为:接种量4%、装液量200 mL/1000 mL、发酵时间6 d、发酵温度28℃、转速220 r/min.在此优化条件下,morindolestatin的产量为2.1799±0.0333 mg/L,比初始含量提高196%,表明发酵条件优化效果显著.
Morindolestatin,a type of indole alkaloid,is derived from Streptomyces sp.HUZ0001 and has a unique structure with excellent antibacterial and antioxidant activities.To increase the production of morindolestatin,a combination of single factor experiments and orthogonal design experiments was conducted to optimize the fermentation conditions,including the type of medium,fermentation time,inoculum size,liquid volume,fermentation temperature,and rotation speed,using Streptomyces sp.HUZ0001 as the test strain.The results showed that the optimized fermentation medium consisted of 2.0%soluble starch,1.0%cottonseed meal powder,0.5%yeast extract,0.5%malt extract,0.2%MgSO_(4)·7H_(2)O,0.2%NaCl,and 0.2%CaCO_(3).The optimized fermentation conditions were an inoculum size of 4%,a liquid volume of 200 mL/1000 mL,a fermentation time of 6 days,a fermentation temperature of 28℃,and a rotation speed of 220 r/min.Under these optimized conditions,the yield of morindolestatin was 2.1799±0.0333 mg/L,which was 196%higher than the initial content,indicating a significant improvement in the fermentation conditions.
