摘要 目的 :分离、培养和鉴定胎鼠脊髓神经干细胞 ,为建立稳定的脊髓神经干细胞系提供基础。方法 :应用无血清培养技术从胎鼠脊髓分离、培养神经干细胞 ,并应用神经干细胞特异性抗体对其进行标记 ;应用 Brd U掺入培养检测其分裂增殖能力 ;应用神经元和星形胶质细胞特异性抗体分析其多向分化潜能。结果 :分离到的细胞具有不断分裂增殖的能力 ,可被神经干细胞特异性抗体所标记 ,并在血清条件下分裂为神经系统多种细胞。结论 :分离到的细胞具有自我复制能力和多向分化潜能 ,是中枢神经系统的干细胞 。 Objective:To isolate, cultivate,and identify the neural stem cells from fetal rat spinal cord, and provide the basis for the set up of spinal cord NSC lineage and the appliance to experimental spinal cord injury or brachial plexus root avulsion. Methods:The neural stem cells were isolated and cultivated using serum free culture techniques. The primary cells and differentiated cells were identified by specific antibodies,respectively. The potential of division and expansion of the cells were analyzed by culture within medium with Brdu. Results:The isolated cells can divide ,expand ,and were identified with NSC specific antibody the cells.Cultured within the medium containing fetal bovine serum can differentiate into multiple types of cells of CNS. Conclusion:The isolated cells can self renew and have multipotential.Those cells were neural stem cells of central nervous system.
