摘要
目的观察电针神庭、百会穴后对缺血再灌注大鼠学习记忆能力的改善情况及海马区哺乳动物雷帕霉素靶蛋白(mTOR)表达的影响。方法将符合标准的大鼠按照随机数字表随机分为假手术组、模型组和电针组各10只,采用改良线栓法制备左侧大脑中动脉缺血再灌注大鼠模型,造模后2 h和第14天分别采用Zea-Longa评分评估各组神经功能缺损情况。电针组给予电针神庭、百会穴治疗14 d,1次/d,30 min/次,模型组和假手术组只在同等条件下抓取。通过Morris水迷宫实验检测大鼠的行为学,通过反转录(RT)-聚合酶链反应(PCR)检测各组海马区mTOR mRNA表达。结果治疗前及治疗后,与假手术组比较,模型组及电针组Longa评分显著升高(P<0.05);与模型组比较,电针组Longa评分显著降低(均P<0.05)。与假手术组比较,模型组及电针组第9~13天逃避潜伏期显著增加、穿越逃生平台次数显著减少(P<0.05);与模型组比较,电针组第9~13天逃避潜伏期显著减少、穿越逃生平台次数显著增加(均P<0.05)。与模型组相比,电针组mTOR表达显著增高,假手术组mTOR表达显著降低(均P<0.05)。结论电针神庭、百会穴在改善脑缺血再灌注大鼠的学习及记忆能力方面疗效显著,且可通过调控脑缺血再灌注后损伤大鼠海马区自噬相关蛋白mTOR表达以保护大鼠脑组织。
Objective To observe the effect of electroacupuncture at Shenting and Baihui points on the improvement of learning and memory ability and the expression of mammalian target of rapamycin(mTOR)in hippocampus of ischemia-reperfusion rats.Methods The rats meeting the criteria were randomly divided into sham operation group,model group and electroacupuncture group according to random number table method,with 10 rats in each group.The left middle cerebral artery ischemia-reperfusion rat model was prepared by modified thread thrombus method.The neurological impairment of each group was evaluated by Zea-Longa score at 2 h and 14 d after modeling.The electroacupuncture group was treated with electroacupuncture at Shenting and Baihui points for 14 d,once per day for 30 min each time,and the model group and the sham operation group were only captured under the same conditions.The behavior of rats was detected by Morris water maze test,and the expression of mTOR mRNA in hippocampus of each group was detected by RT-polymerase chain reaction(PCR).Results Before and after treatment,Longa score in model group and electroacupuncture group was significantly higher than that in sham operation group(P<005);compared with model group,Longa score in electroacupuncture group was significantly decreased(all P<0.05).Compared with the sham operation group,the escape latency of the model group and the electroacupuncture group was significantly increased and the number of crossing the escape platform was significantly decreased on day 9th to 13th;compared with the model group,the escape incubation period was significantly decreased and the number of crossing escape platform was significantly increased in the electroacupuncture group on day 9th to 13th(all P<0.05).Compared with the model group,the expression of mTOR in electroacupuncture group was significantly increased,while that in sham operation group was significantly decreased(all P<0.05).Conclusions Electroacupuncture at Shenting and Baihui points could significantly improve the learning and memory ability of cerebral ischemia-reperfusion rats,and could protect the brain tissue of rats by regulating the expression of autophagy related protein mTOR in hippocampus of rats injured by cerebral ischemia-reperfusion.
