乐果和苦参碱农药在柞蚕幼虫体内的代谢研究

摘要 为了探明乐果和苦参碱农药在柞蚕幼虫体内的代谢情况,以5龄眠起第3天柞蚕幼虫为试虫,乐果和苦参碱均设置高、低两个剂量组,利用浸叶法对试虫进行经口一次性染毒,利用高效液相色谱-串联质谱法检测染毒后不同时间内试虫体内乐果和苦参碱的浓度,通过非房室模型统计矩参数,计算药物在试虫体内的消除半衰期。动力学非房室模型统计矩参数表明,低、高剂量乐果的Cmax分别为3075μg/kg和5 359.8μg/kg, Tmax分别为2 h和1 h, AUC(0-t)分别为34 060.292μg/(L·h)和18 505.95μg/(L·h),t1/2β分别为32.764 h和36.551 h;低、高剂量苦参碱的Cmax分别为0.443μg/kg和0.281μg/kg, Tmax均为1 h, AUC((0-t))分别为0.868μg/(L·h)和4.668μg/(L·h),t1/2β分别为0.723 h和27.239 h。试验表明:低剂量乐果农药在试虫体内驻留能力强,衰减速度慢,而高剂量则相反;高、低剂量苦参碱农药均能快速被蚕体吸收和分布,但是高剂量的苦参碱农药在试虫体内存留能力强、持续作用时间长。 The aim of this study was to investigate the metabolism of Dimethoate and Matrine pesticides in tussah larvae.The larvae of tussah on the third day after the pentamolter were used as the test insects.Both Dimethoate and Matrine were administered to the test insects in two dose groups,high and low,through a one-time oral intoxication using the leaf dipping method.The concentrations of Dimethoate and Matrine in the test insects were measured at different times after poisoning using HPLC-MS/MS,and the half-life of elimination in the test insects was calculated using the statistical moment parameter of the non-compartmental model.Statistical moment parameters of the kinetic non-compartment model showed that the C max of low-dose group and high-dose group of Dimethoate were 3075μg/kg and 5359.8μg/kg,respectively,with a T max of 2 h and 1 h.The AUC(0-t)for both groups were 34060.292μg/(L·h)and 18505.95μg/(L·h),respectively,with a t 1/2βof 32.764 h and 36.551 h.The C max of low-dose group and high-dose group of Matrine were 0.443μg/kg and 0.281μg/kg,respectively,and the T max for both groups was 1 hour.The AUC(0-t)was 0.868μg/(L·h)and 4.668μg/(L·h),and the t 1/2βwas 0.723 h and 27.239 h for the low-dose and high-dose groups,respectively.It is concluded that the low-dose group of Dimethoate pesticides demonstrated a strong ability to reside in the body of test insects and decay slowly,while the high-dose group showed the opposite pattern;high and low dose groups of Matrine pesticides were rapidly absorbed and distributed by tussah larvae,but the high-dose group had a stronger ability to reside in the body of test insects and showed a longer duration of action.
